宫颈癌组织基因表达谱差异筛选及功能预测 被引量:1

Differential screening and functional prediction of gene expression profiles in cervical cancer tissues

摘要 目的筛选宫颈癌组织中的差异表达mRNA,并探讨其生物学功能以及参与的信号通路。方法通过对微阵列数据库GSE63514和GSE55940中的33例宫颈癌患者的宫颈癌组织和癌旁组织基因芯片数据进行生物信息学分析,筛选出差异表达mRNA,并预测其参与的生物学功能和信号通路。结果宫颈癌组织中,98个mRNA上调,138个mRNA下调;其中CTHRC1、ZIC2、MMP12、AIM2、CA9的表达显著上调,CRNN、CRISP3、TMPRSS11B、SPINK7、MAL的表达显著下调(P<0.05)。基因本体论(GO)富集分析结果显示,表达上调的mRNA涉及220个生物学过程,表达下调的mRNA涉及83个生物学过程。京都基因与基因组百科全书(KEGG)通路富集分析显示,上调mRNA显著富集到Cell cycle、DNA replication、Toll-like receptor signaling pathway、Bladder cancer和IL-17 signaling pathway 5条信号通路,下调mRNA显著富集到Arachidonic acid metabolism和Staphylococcus aureus infection 2条信号通路。结论本研究得到了宫颈癌组织中mRNA表达谱,其中异常表达的mRNA(CTHRC1和CRNN)可能在宫颈癌的生理病理学中发挥关键作用。 Objective To screen differential expression mRNA in cervical cancer tissues,and to explore its biological functions and involved signal pathways.Methods Bioinformatics analysis was carried out on gene chip data of 33 cervical cancer patients'cervical cancer tissues and adjacent tissues in the microarray databases GSE63514 and GSE55940.Differential expression mRNA was screened,and its biological functions and signal pathways were predicted.Results Ninety-eight mRNA were up-regulated and 138 mRNA were down-regulated in cervical cancer tissues;the expression of CTHRC1,ZIC2,MMP12,AIM2 and CA9 were significantly up-regulated,while the expression of CRNN,CRISP3,TMPRSS11B,SPINK7 and MAL were significantly down-regulated(P<0.05).The results of Gene Ontology(GO)enrichment analysis showed that the mRNA with up-regulated expression involved 220 biological processes and the mRNA with down-regulated expression involved 83 biological processes.Enrichment analysis of Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway showed that up-regulated mRNA are significantly enriched to 5 signal pathways,including Cell cycle,DNA replication,Toll-like receptor signaling pathway,Bladder cancer and IL-17 signaling pathway,and down-regulated mRNA are significantly enriched to 2 signal pathways,including Arachidonic acid metropolism and Staphylococcus aureus infection.Conclusion This study has obtained the mRNA expression profile in cervical cancer tissues,and the abnormally expressed mRNA(CTHRC1 and CRNN)may play key roles in the physiology and pathology of cervical cancer.
