正清风痛宁治疗神经病理性疼痛的动物模型研究

摘要 目的:观察正清风痛宁对坐骨神经慢性缩窄性损伤(Chronic constriction injury,CCI)致小鼠神经病理性疼痛的缓解作用,并探讨其部分作用机制。方法:将30只C57BL/6小鼠随机分成3组,实验组(建模后给予正清风痛宁)、模型对照组(建模后给予生理盐水)、假手术组(术后给予生理盐水)。建立小鼠神经病理性疼痛动物模型。比较三组术前及建立神经病理性疼痛模型后(术后)1、3、5、7、10、14 d小鼠的机械缩足反射阈值(paw withdrawal mechanical threshold,PWMT)、热刺激缩足反应潜伏期(paw withdrawal thermal latency,PWTL)。术后14 d处死各组小鼠,ELISA检测各组小鼠脊髓组织肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)等炎症因子。结果:从CCI术后1 d开始,模型对照组小鼠的PWMT明显低于假手术组,PWTL明显短于假手术组(P<0.05)。相比于模型对照组,术后第3天起给予正清风痛宁40 mg/(kg·d)后,从术后7 d起实验组小鼠的PWMT明显升高,PWTL明显延长(P<0.05)。术后14 d,模型对照组小鼠脊髓中的TNF-α、IL-1β和IL-6表达水平均明显高于假手术组(P<0.05),相比于模型对照组,实验组中TNF-α、IL-1β和IL-6的表达水平均降低(P<0.05)。结论:正清风痛宁对CCI致小鼠神经病理性疼痛有明显的缓解作用,其机制可能与其下调TNF-α、IL-1β、IL-6相关。 Objective:To observe the effect of Zhengqing Fengtongning on Chronic sciatic nerve constriction injury (CCI) induced neuropathic pain in mice and to explore its mechanism.Method:A total of 30 C57BL/6 mice were randomly divided into 3 groups:experimental group (given Zhengqing Fengtongning after modeling),model control group (given normal saline after modeling),and sham operation group (given normal saline after operation).The animal model of neuropathic pain in mice were established.The paw withdrawal mechanical threshold (PWMT) and paw withdrawal thermal latency (PWTL) of mice before operation and 1,3,5,7,10,and 14 d after the establishment of neuropathic pain model (after operation) were compared among three group.The mice in each group were sacrificed 14 days after operation,inflammatory factors such as tumor necrosis factor-α (TNF-α),interleukin-1β (IL-1β) and interleukin-6 (IL-6) were detected by ELISA.Result:From 1 day after CCI,PWMT of the model control group were significantly lower than that of the sham operation group,and PWTL were significantly shorter than that of the sham operation group (P<0.05).Compared with the model control group,the PWMT and PWTL of 7 d after operation of the experimental group were significantly increased and prolonged after the treatment with Zhengqing Fengtongning 40 mg/ (kg·d) from the third day after operation (P<0.05).14 d after operation,the expression levels of TNF-α,IL-1β and IL-6 in the spinal cord of mice in model control group were significantly higher than those in sham operation group (P<0.05),compared with the model control group,the expression levels of TNF-α,IL-1β and IL-6 in the experimental group were decreased (P<0.05).Conclusion:Zhengqing Fengtongning can relieve neuropathic pain induced by CCI in mice,and its mechanism may be related to the down-regulation of TNF-α,IL-1β,IL-6.
