摘要
目的研究白细胞介素33(interleukin-33,IL-33)对心肌梗死(myocardial infarction,MI)后小鼠骨髓巨核细胞增殖、生成血小板的影响。方法将20只C57BL6雄性小鼠随机分为两组,即对照组和IL-33处理组。对照组腹腔注射PBS连续14天,然后结扎小鼠冠脉前降支造成MI。IL-33处理组腹腔注射IL-33蛋白(50μg/kg)连续14天,然后如前进行MI造模。MI 3天后,采用全血细胞计数仪检测小鼠循环血液中血小板计数;采用免疫荧光染色与流式细胞术检测小鼠骨髓中成熟巨核细胞、未成熟巨核细胞以及血小板前体数量。结果经IL-33处理后,小鼠骨髓中巨核细胞与巨核祖细胞的数量增多,然而血小板前体数量与循环血液中血小板水平比较,差异无统计学意义,血清血小板生成素(thrombopoietin,TPO)水平亦无明显变化。结论IL-33促进心肌梗死后骨髓巨核细胞增殖,但不影响血小板生成。
Objective To study the effect of interleukin-33(IL-33)on the proliferation and platelet production of bone marrow megakaryocytes in mice after myocardial infarction.Methods Twenty C57BL6 male mice were randomly divided into two groups:control group and IL-33 treated group.In control group,PBS was injected intraperitoneally for 14days,and then the anterior descending coronary artery of mice was ligated to induce MI.In IL-33 treated group,IL-33 protein was injected intraperitoneally(50μg/kg)for 14 consecutive days,and then MI was induced as described above.Three days after MI,the number of platelets in the peripheral blood of mice was detected by whole blood cell counter.Immunofluorescence staining and flow cytometry were used to detect the number of mature megakaryocytes,immature megakaryocytes and proplatelet in the mouse bone marrow.Results After IL-33 treatment,the number of mature megakaryocytes and immature megakaryocytes in mouse bone marrow increased.There was no significant difference in the number of proplatelet per image area and the level of platelets in the circulating blood,and there was no significant change in serum TPO level between control group and IL-33 treated group.Conclusion IL-33 promotes the proliferation of bone marrow megakaryocytes after myocardial infarction,but does not affect platelet production.
