基于UPLC-MS/MS广靶代谢组学技术分析不同陈化年限广陈皮化学成分的差异 被引量:5

摘要 目的 研究不同陈化年限广陈皮中化合物的差异,为广陈皮陈化机制提供依据。方法 运用超高效液相色谱串联质谱(UPLC-MS/MS)广泛靶向技术对3种陈化年限(1 a、3 a和5 a)的广陈皮化合物进行检测,使用主成分分析和偏最小二乘判别分析法(PLS-DA)鉴定差异代谢物。结果 共检出562个化合物,主要成分为黄酮类(109个)和苷类(86个)等成分,涉及不饱和脂肪酸的生物合成、黄酮类生物合成、黄酮和黄酮醇的生物合成等14条代谢通路。PLS-DA筛选出175个差异化合物,其中陈化5 a组与1 a组比较差异代谢物最多有126个,陈化3 a组与1 a组比较差异代谢物有105个,陈化5 a组与3 a组比较差异代谢物最少,仅41个,表现出陈化1~5 a差异化学成分含量以增加为主,陈化3~5 a差异化学成分呈现减少的趋势。结论 广陈皮在3~5 a陈化期间化学成分有一个相对多的阶段,这为广陈皮陈化3 a以上使用为佳提供了理论基础。 Objective To study the differences of compounds in Citri reticulatae Pericarpium “ Chachi” with different aging years, and to provide the basis for the aging mechanism of Citri reticulatae Pericarpium “ Chachi”.Methods Ultra-high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) broadtargeted technology was used to detect 3 kinds of Citri reticulatae Pericarpium “ Chachi” compounds aged one year, three years and five years, and the differential metabolites were identified by principal component analysis and partial least squares analysis(PLS-DA). Results A total of 562 compounds were detected, the main components were flavonoids(109) and glycosides(86), and 14 metabolic pathways were involved in biosynthesis of unsaturated fatty acids, biosynthesis of flavonoids, and biosynthesis of flavonoids and flavonols. 175differential compounds were screened by PLS-DA, among which there were 126 differential metabolites between the 5-year aging group and 1-year aging group, and 105 differential metabolites between the 3-year aging group and 1-year aging group, between the 5-year aging group and the 3-year aging group, the number of differential metabolites was only 41 which is the least. The content of the differential chemical components increased mainly during the 1-5 a of aging, and the differential chemical components of the 3-5 a aging showed a decreasing trend. Conclusion The chemical composition of Citri reticulatae Pericarpium “ Chachi” has a relatively large number of stages during the aging period of 3 to 5 a, which provides a theoretical basis for the optimal use of Citri reticulatae Pericarpium “ Chachi” for more than 3 a of aging.
