摘要
目的:建立HPLC法同时测定山地虎耳草中獐牙菜苦苷、绿原酸、当药苷、金丝桃苷、当药醇苷含量的方法。方法:采用Waters SunFire C_(18)柱(4.6 mm×150 mm,5μm),以甲醇-水(含0.05%磷酸)为流动相,流速为1.0 ml·min^(-1),检测波长为254 nm,柱温为30℃。结果:獐牙菜苦苷、绿原酸、当药苷、金丝桃苷、当药醇苷进样质量分别在0.0506~0.6072μg(r=0.9999)、0.1134~1.3608μg(r=0.9998)、0.0198~0.2382μg(r=0.9998)、0.1182~1.4184μg(r=0.9999)、0.0450~0.5397μg(r=0.9999)范围内呈良好的线性关系,加样回收率分别为93.85%,93.10%,97.22%,98.74%,96.55%,RSD分别为1.02%,1.77%,1.60%,0.69%,1.18%(n=6)。结论:建立的高效液相色谱法简便、高效、准确、经济实用,具有良好的重复性和稳定性,为山地虎耳草的质量评价,以及综合利用开发药用资源提供了重要参考依据。
Objective:To establish an HPLC method to detect swertiamarin,chlorogenic acid,chiratin,hypericin and swertianolin in Saxifraga montana.Methods:A Waters SunFire C_(18) column(4.6 mm×150 mm,5μm)was used with the mobile phase of methanol-water(containing 0.05%phosphoric acid).The flow rate was 1.0 ml·min^(-1),the detection wavelength was set at 254 nm,and the column temperature was set at 30℃.Results:The five constituents showed good linear relationships within the range of 0.0506-0.6072μg(r=0.9999)for swertiamarin,0.1134-1.3608μg(r=0.9998)for chlorogenic acid,0.0198-0.2382μg(r=0.9998)for chiratin,0.1182-1.4184μg(r=0.9999)for hypericin,and 0.0450-0.5397μg(r=0.9999)for swertianolin.The average recoveries of five constituents were 93.85%,93.10%,97.22%,98.74%and 96.55%,and the RSDs were 1.02%,1.77%,1.60%,0.69%and 1.18%(n=6),respectivly.Conclusion:The established HPLC method is simple,efficient,accurate,economical and practical with good repeatability and stability,which provids important reference for the quality evaluation of Saxifraga montana and the comprehensive utilization and development of medicinal resources.
