高粱泡叶化学成分的UHPLC-Q Exactive Focus MS/MS分析

Chemical Composition Analysis of Rubus Lambertianus Ser Leaves Based on UPLC-Q-Exactive Focus-MS/MS and Network Pharmacology on Its Anticancer Mechanism

摘要 目的采用超高效液相色谱-四极杆/静电场轨道阱高分辨质谱法(UHPLC-Q Exactive Focus MS/MS)对高粱泡叶的化学成分进行快速识别和鉴定。方法选择高粱泡叶的80%乙醇提取液作为供试品溶液,采用ACE Ultracore2.5 SuperC18(100 mm×2.1 mm,0.5μm)色谱柱(2.1 mm×150 mm,2.6μm),流动相乙腈(C)-0.1%甲酸水溶液(B)梯度洗脱,流速0.3 mL/min,柱温40℃;质谱分析采用加热电喷雾离子源(HESI),正、负离子模式扫描,扫描范围100~1500 m/z。结果共鉴定出了61个成分,包括游离氨基酸、酚酸类、黄酮类、有机酸类、糖苷类及其他类。结论该方法可系统、准确、快速地对高粱泡叶中化学成分进行定性分析,可为高粱泡叶药效物质研究及质量评价提供重要依据。 Objective To rapidly identify the chemical constituents of Rubus lambertianus Ser leaves by ultra-high performance liquid chromatography-quadrupole/electrostatic field orbitrap high resolution mass spectrometry(UHPLC-Q Exactive Focus MS/MS).Methods The 80%ethanol extract of Rubus lambertianus Ser leaves was selected as the test solution.The ACE Ultracore2.5 SuperC18(100 mm×2.1 mm,0.5μm)column(2.1 mm×150 mm,2.6μm)was used.The mobile phase was acetonitrile(C)-0.1%formic acid aqueous solution(B)gradient elution,the flow rate was 0.3 mL/min,and the column temperature was 40℃.Mass spectrometry analysis was performed using a heated electrospray ionization(HESI)source with positive and negative ion mode scanning in the range of 100~1500 m/z.Results A total of 61 components were identified,including free amino acids,phenolic acids,flavonoids,organic acids,glycosides and others.Conclusion:This method can systematically,accurately and quickly analyze the chemical components in sorghum leaves,which can provide an important basis for the study and quality evaluation of Rubus lambertianus Ser leaves.
