姜黄素对AIH小鼠肠道菌群失衡的改善作用

摘要 目的 探讨姜黄素(curcumin, Cur)对自身免疫性肝炎(autoimmune hepatitis, AIH)小鼠肠道菌群失衡的改善作用。方法 BALB/c小鼠随机分成空白对照组、AIH模型组、姜黄素组和地塞米松(阳性药)组。采用刀豆蛋白A(concanavalin A,ConA)尾静脉注射建立小鼠AIH模型。收集小鼠回盲部粪便进行DNA抽提和PCR扩增获取碱基序列,并建立数据库;探索肠道菌群与自身免疫性肝炎的相关性及姜黄素对其肠道菌群失衡的影响。结果 与空白对照组比,AIH模型组小鼠粪便中肠道菌群中拟杆菌属(Bacteroides)、葡萄球菌属(Staphylococcus)丰度明显下降,而norank-f-Muribaculaceae、毛螺旋菌属(Lachnospiraceae-NK4A136-group)、脱硫弧菌属(Desulfovibrio)、罗斯氏菌属(Roseburia)、Jeotgalicoccus、糖化假丝酵母菌(Candidatus-Saccharimonas)、Lachnoclostridium、厌氧棍状菌属(Anaerotruncus)丰度有所上升;与AIH模型组比较,姜黄素组和地塞米松组中葡萄球菌属(Staphylococcus)、norank-f-Muribaculaceae、毛螺旋菌属(Lachnospiraceae-NK4A136-group)、Jeotgalicoccus、棒杆菌属(Corynebacterium)、芽孢八叠球菌属(Sporosarcina)丰度有所下降,而拟杆菌属(Bacteroides)、乳酸菌属(Lactobacillus)、毛螺旋菌属(norank-f-Lachnospiraceae)、Colidextribacter、细小杆菌(Parvibacter)、Harryflintia丰度有所上升。结论 与正常小鼠相比,AIH小鼠肠道菌群结构发生改变,且姜黄素对AIH小鼠的肠道菌群失衡有明显改善作用。 Objective To explore the effect of curcumin on intestinal flora imbalance in autoimmune hepatitis(AIH) mice. Methods BALB/c mice were randomly divided into 4 groups: blank control group, AIH model group, curcumin group and dexamethasone(DXMS) group. The model of AIH mice was established by tail vein injection of Concanavalin A(ConA). We collected feces from ileocecal region of mice for DNA extraction and obtained the base sequences of feces by PCR amplification, and then established a database. The correlation between intestinal flora and autoimmune hepatitis was observed and the effect of curcumin on the imbalance of intestinal flora was analyzed. Results Compared with those of the blank control group, the abundance of Bacteroides and Staphylococcus in the feces of the model group decreased significantly. However, the abundance of norank-f-Muribaculaceae,Lachnospiraceae-NK4A136-group, Desulfovibrio,Roseburia,Jeotgalicoccus,Candida Saccharimonas,Lachnoclostridium and Anaerotruncus increased. Compared with those of the model group, the abundance of Staphylococcus,norank-f-Muribaculaceae,Lachnospiraceae-NK4A136-group,Jeotgalicoccus,Corynebacterium and Sporosarcina decreased in curcumin group and dexamethasone group and the abundance of Bacteroides,Lactobacillus,norank-f-Lachnospiraceae,Colidextribacter,Parvibacter and Haryflintia increased. Conclusion Compared with that of the blank control group, the structure of intestinal flora changed in AIH mice. Curcumin can improve the imbalance of intestinal flora in mice with autoimmune hepatitis.
