摘要
目的 比较中药材桔梗科植物党参和桔梗的基因组大小。方法 分别摘取党参和桔梗的新鲜嫩叶为实验材料,以蒙古黄芪作为党参的内标,以大豆作为桔梗的内标,采用裂解液(lysis buffer, LB)01制备各细胞核悬液,碘化丙啶染色,用流式细胞仪分别测定党参和桔梗的荧光强度。结果 估测出党参的基因组大小约为1.00 Gb;桔梗的基因组大小约为0.70 Gb。结论 桔梗科中药材党参基因组大小大于桔梗的基因组大小,相差约0.30 Gb,为同科的党参和桔梗之间的差异提供了重要支持。
Objective To investigate the genome sizes of traditional medicinal herbs Dangshen[Codonopsis pilosula(Franch.) Nannf.]and Jiegeng[Platycodon grandiflorum(Jacq.) A. DC.]from Campanulaceae. Methods The fresh leaves of Dangshen[Codonopsis pilosula(Franch.) Nannf.]and Jiegeng[Platycodon grandiflorum(Jacq.) A. DC.]were picked as the materials of experiment, and Menggu Huangqi[Astragalus membranaceus(Fisch.) Bge. var. mongolicus(Bge.) Hsiao]was chosen as an internal reference of Dangshen[Codonopsis pilosula(Franch.) Nannf.]and Dadou[Glycine max(Linn.) Merr.]was chosen as an internal reference of Jiegeng[Platycodon grandiflorum(Jacq.) A. DC.]. The nuclear suspension was prepared by lysis buffer and was stained by propidium iodide. Flow cytometry was used to detect the fluorescence intensities of Dangshen[Codonopsis pilosula(Franch.) Nannf.]and Jiegeng[Platycodon grandiflorum(Jacq.) A. DC.]. Results The genome size of Dangshen[Codonopsis pilosula(Franch.) Nannf.]was 1.00 Gb, and that of Jiegeng[Platycodon grandiflorum(Jacq.) A. DC.]was 0.70 Gb. Conclusion The genome sizes of Dangshen[Codonopsis pilosula(Franch.) Nannf.]is larger than that of Jiegeng[Platycodon grandiflorum(Jacq.) A. DC.]and the difference is about 0.30 GB, which provides important support for the differences between Dangshen[Codonopsis pilosula(Franch.) Nannf.]and Jiegeng[Platycodon grandiflorum(Jacq.) A. DC.]in the same family.
