摘要
【目的】建立云南红河州番茄褐色果病的检测技术,为控制其传播和治疗提供支撑。【方法】采用电子显微镜负染色观察、ELISA检测以及外壳蛋白基因序列RT-PCR检测及序列分析,明确云南红河州番茄褐色皱果病的病原。【结果】电子显微镜观察到病毒粒子为长300 nm、直径18 nm的杆状病毒,ELISA检测发现与烟草花叶病毒属的烟草花叶病毒(TMV)和番茄花叶病毒(ToMV)有血清学反应;RT-PCR扩增获得的目的片段与番茄褐色皱纹果病毒的病毒基因相似性达99.81%。【结论】引起云南红河番茄褐色皱纹果病的病原是烟草花叶病毒属的番茄褐色皱果病毒。
【Objective】The detection technology of tomato brown rugose fruit virus in Hoghe Prefecture of Yunnan is established,so as to provide the support for controlling and preventing the virus.【Method】The electron microscope negative staining observation,ELISA testing,and RT-PCR detection and sequence analysis of the coat protein gene sequences are conducted to identify the pathogen causing tomato brown rugose fruit virus in Honghe Prefecture of Yunnan.【Result】Baculovirus with a length of 300 nm and diameter of 18 nm is observed by the electron microscope.ELISA testing shows the diseased fruit has a serological reaction with tobacco mosaic virus(TMV)and tomato mosaic virus(ToMV)of tobamovirus.The target fragment amplified by RT-PCR has a high similarity(99.81%)of virus gene with tomato brown rugose fruit virus.【Conclusion】The pathogen which is caused the tomato brown rugose fruit disease in Honghe Prefecture of Yunnan is the tomato brown rugose fruit virus of tobamovirus.
