摘要
目的:探讨姜黄素在类风湿关节炎(RA)患者外周血CD4^(+)T细胞培养中对Th17/Treg细胞平衡的调节作用。方法:从30个未经治疗的RA患者和30个健康受试者的血液中采集CD4^(+)T细胞,用Th17分化因子刺激,用1μg/mL的姜黄素孵化。72 h后,收集细胞并用流式细胞术检测Th17和Treg细胞百分比,用ELISA技术检测IL⁃17和TGF⁃β1水平。结果:与对照组(0μg/mL姜黄素处理组)比较,在RA患者CD4^(+)T细胞中,1μg/mL姜黄素可以降低Th17细胞百分比(P<0.05),减少IL⁃17的产生(P<0.05),但能升高Treg细胞百分比(P<0.01),并增加TGF⁃β1产生(P<0.01)。然而,在健康受试者的CD4^(+)T细胞培养中没有观察到这种现象。结论:姜黄素可以调节RA患者CD4^(+)T细胞中Th17/Treg细胞的平衡,但对健康受试者没有影响。
Objective:To investigate the regulatory effect of curcumin on Th17/Treg cell balance in CD4^(+)T cell culture of rheumatoid arthritis(RA)patients.Methods:CD4^(+)T cells were collected from the blood of 30 untreated RA patients and 30 healthy subjects.These cells were stimulated with Th17 dif⁃ferentiation factors and incubated with 1μg/mL curcumin.After 72 hours,the cells were collected and the percentage of Th17 and Treg cells was measured by flow cytometry.IL⁃17 and TGF⁃β1 levels were detected by ELISA.Results:In the CD4^(+)T cells of RA patients,as compared with con⁃trol group,curcumin(1μg/mL)reduced the percentage of Th17 cells(P<0.05)and the production of IL⁃17(P<0.05),but increased the percentage of Treg cells(P<0.01)and TGF⁃β1 production(P<0.01).However,these changes were not found in CD4^(+)T cell cultures in healthy subjects.Conclusion:These data suggest that curcumin can modulate Th17/Treg cell balance of CD4^(+)T cells in RA patients and has no effect on CD4^(+)T cells in healthy subjects.
