帕勒隆尼氏假单孢菌HG-258发酵条件和培养基的优化

摘要 对HG-258菌株的生长特性进行了研究,并对其发酵条件及培养基进行了优化。结果表明,HG-258菌株的生长规律:0~4 h延滞期,4~24 h对数生长期,24~36 h稳定期,36 h以后衰亡期;最佳发酵条件:初始pH 7.0,培养温度28℃,接种量3%,转速200 r/min;最适培养基:蔗糖10.0 g/L,甘油10.0 g/L,酵母膏30.0 g/L,豆粕10.0 g/L,硫酸镁0.1 g/L,磷酸二氢钾0.05 g/L,在最佳发酵条件下,摇瓶发酵液菌数可达2.826×10^(10)CFU/mL,相比初始培养基及培养条件下的发酵水平提高了约1倍。 In this study,the growth characteristics of P.palleroniana strain HG-258 was studied and the fermentation conditions and media for HG-258 were optimized.The results showed that the growth law of HG-258 was:0-4 h lag period,4-24 h logarithmic growth period,24-36 h stable period,decline phase ofter 36 h.The optimized condition for the fermentation of HG-258 were as followings:starting pH 7.0,fermentation temperature 28℃,the rate of inoculum 3%,speed of rotation 200 r/min.The composition of optimized medium was sucrose 10.0 g/L,glycerol 10.0 g/L,yeast extract 30.0 g/L,soybean flour 10.0 g/L,Magnesium sulphate 0.1 g/L,potassium dihydrophosphate 0.05 g/L.Under the optimized condition,the cell count of shake-flask fermentation culture reached 2.826×10^(10) CFU/mL with the optimized medium,which was about one times higher than that with the starting medium and fermentation condition.
