1株野生灵芝的分离鉴定及生物学特性分析 被引量:5

摘要 为开发南京紫金山野生生物资源,对采自紫金山的一株野生菌株ZJ-1进行了分离鉴定。采用形态学观察和ITS序列分析相结合的方法鉴定该野生菌株为灵芝(Ganoderma lucidum),并对菌株ZJ-1开展了菌丝培养特性研究及驯化栽培试验。结果表明其最适生长温度为28℃~32℃,适宜pH为4.0~5.0,最适碳源为可溶性淀粉,最适氮源为酵母粉;通过人工栽培成功获得了灵芝菌株ZJ-1的子实体。利用扫描电镜发现灵芝菌管内有着规则的网状结构,其上布满卵形孢子。研究结果将为菌株ZJ-1的开发利用提供数据参考。 In order to develop the wild resources of Zijin Mountain in Nanjing,the wild strain ZJ-1 collected from Zijin Mountain was isolated and identified.It was identified as Ganoderma lucidum through morphological observation and ITS sequence analysis.The mycelial culture characteristics and domesticated cultivation experiments of strain ZJ-1 were carried out.The results showed that the optimum growth temperature was 28℃-32℃,the optimum pH was 4.0-5.0,the optimum carbon source was soluble starch,and the optimum nitrogen source was yeast extract.The fruit bodies of G.lucidum strain ZJ-1 were successfully obtained in the artificial cultivation.Observation under the scanning electron microscope showed that the tube of G.lucidum had a regular network structure,full of ovoid spores.Results of this research will provide a data reference for the development and utilization of strain ZJ-1.
