摘要
转录因子B-box(BBX)属于锌指蛋白家族的一个亚家族,在植物生长、发育和非生物胁迫响应途径中表现出多种生物学功能。该研究根据基于隐马尔可夫模型(HMM)的图谱(B-box型锌指结构域,PF00643),使用SMART和Pfam数据库筛选和验证了银杏(Ginkgo biloba)全基因组注释蛋白质,从中鉴定出12个GbBBX基因,并对GbBBX蛋白成员的保守结构域、基因结构、系统发育关系和组织表达谱进行了分析。结果表明:GbBBX蛋白家族包含1~2个保守的B-box型锌指蛋白结构域和1个CCT型结构域;GbBBX基因家族的外显子数量在1~5个之间,75%的家族成员外显子数量为3或5个;与拟南芥和番茄的BBX蛋白构建的邻接(NJ)系统发育树显示,大部分的GbBBX蛋白被聚集在进化枝I中,而拟南芥和番茄的BBX蛋白聚集在其他4个进化枝中;GbBBX在植物发育、激素和胁迫反应相关途径中富集;GbBBX基因在各组织中组成型表达,推测GbBBX基因可能参与了银杏各组织的生长发育调控。
The transcription factor B-box(BBX)belongs to the zinc finger protein subfamily and can be involved in a variety of pathways regulating plant growth,development and abiotic stress response.In this study,12 GbBBX genes were identified using SMART and Pfam databases based on hidden Markov model(HMM)maps(B-box-type zinc finger domains,PF00643)and the conserved domain,gene structure,phylogenetic relationships and tissue expression profiles of GbBBX protein members were analyzed.The results showed that:the GbBBX protein family contains 1-2 conserved B-box-type zinc finger domains and one CCT-type domain;the number of exons in the GbBBX gene family ranged from one to five,with 75%of the family members having three or five exons;the neighbor-joining(NJ)phylogenetic tree constructed with the BBX proteins of Arabidopsis thaliana and Solanum lycopersicum showed that most of the GbBBX proteins were clustered in clade I,while BBX proteins from Arabidopsis thaliana and Solanum lycopersicum were clustered in the other four clades;GbBBX was enriched in pathways related to plant development,hormone and stress responses;GbBBX genes were constitutively expressed in various tissues,indicating that GbBBX genes might be involved in the regulation of growth and development in various tissues of Ginkgo biloba.
