摘要
目的考察参附注射液对小鼠脑缺血再灌注损伤后热休克蛋白27(HSP27)的表达以及NF-κB信号通路的影响。方法将C57/BL小鼠随机分为假手术组,模型组,参附注射液低、中、高剂量组(2.5、5、10 mL/kg),采用线栓法制备小鼠脑缺血再灌注模型,造模时给药,缺血1h再灌注24 h后,测定各组小鼠神经功能评分、脑梗死体积,观察大脑病理形态变化。采用免疫组织化学检测脑组织中HSP27蛋白表达情况,及对核因子-κB(NF-κB)信号通路的影响。结果与模型组相比,参附注射液能改善给药组小鼠神经功能损伤症状、降低小鼠脑梗死体积以及改善脑组织病理形态。同时上调小鼠脑组织中HSP27蛋白的表达,并抑制NF-κB信号通路的激活。结论参附注射液对小鼠脑缺血再灌注损伤具有一定的改善作用,其作用机制可能与上调HSP27蛋白表达和抑制NF-κB信号通路激活有关。
Objective To investigate the effect of Shenfu injection on the expression of heat shock protein 27(HSP27) and NF-κB pathway after cerebral ischemia-reperfusion injury in mice. Methods C57/BL mice were randomly divided into sham group, model group, low, middle and high dose group(2.5,5,10 mL/kg) of Shenfu injection, and underwent right middle cerebral artery occlusion for 1 hour with a subsequent 24-hour reperfusion to produce I/R injury. Shenfu injection was then given intraperitoneally. The neurological function score, cerebral infarction volume and the pathological changes of brain in mice were observed. The expression of HSP27 protein and NF-κB pathway was detected by immunohistochemistry in brain tissue. Results Compared with the model group, Shenfu injection can improve the symptoms of neurological injury, reduce the volume of cerebral infarction and the pathological morphology of brain tissue in the administration group. Meanwhile, it can increase the expression of HSP27 protein and inhibit the activation of NF-κB pathway in mice.Conclusion Shenfu injection can improve cerebral ischemia-reperfusion injury in mice, and its mechanism may be related to up-regulating the expression of HSP27 protein and inhibiting NF-κB signal pathway activation.
