高脂血症脾虚痰浊猪血清高密度脂蛋白向失功能性高密度脂蛋白转变 被引量:2

Change of Serum High Density Lipoprotein to Dysfunctional High Density Lipoprotein in Small Pig Model of Hyperlipidemia with Spleen Deficiency and Phlegm-Turbid Syndrome

摘要 目的探讨高脂血症脾虚痰浊猪血清高密度脂蛋白(high density lipoprotein,HDL)组成成分变化。方法普通级广西巴马小型猪10只随机分为正常组、模型组,每组5只。正常组给予普通饲料喂养,模型组给予高脂饲料喂养24周建立高脂血症脾虚痰浊证小型猪模型。于第0、24周,经小猪耳静脉采集空腹血样,采用ELISA法检测血清HDL、高密度脂蛋白-载脂蛋白AI(HDL-Apo AI)、高密度脂蛋白-淀粉样蛋白A(HDL-SAA)、高密度脂蛋白-对氧磷酶1(HDL-PON1)水平,并计算HDL-SAA/HDL-ApoAI比值,采用HPLC法测定高密度脂蛋白-1磷脂鞘氨醇(HDL-S1P)的含量。结果实验24周时高脂血症脾虚痰浊猪血清HDL水平与实验0周及同期正常组相比,均降低(P<0.05);且实验24周时高脂血症脾虚痰浊猪血清HDL-ApoAI、HDL-PON1、HDL-S1P水平无论与实验0周相比,还是与同期正常组相比,都是显著降低的(P<0.01);同时,实验24周模型组小猪血清HDL-SAA水平、HDL-SAA/HDL-ApoAI比值则显著升高(P<0.01)。结论高脂血症脾虚痰浊猪血清HDL水平显著降低,组分改变,导致介导胆固醇外流、抗炎、抗氧化作用减弱,变得“失功能”。 Objective To investigate the changes of serum high density lipoprotein(HDL)composition in pigs of hyperlipidemia with spleen deficiency and phlegm-turbid syndrome.Methods A total of 10 ordinary Guangxi Bama miniature pigs were randomly divided into a normal group and a model group,with 5 pigs in each group.The normal group was fed with basic food,and the model group was fed with high-fat food for 24 weeks to establish a mini pig model of hyperlipidemia with spleen deficiency and phlegm-turbid syndrome.At week 0 and 24,fasting blood samples were collected by ear vein.Serum levels of HDL,high density lipoprotein-Apo lipoprotein AI(HDL-apoAI),high density lipoprotein-amyloid A(HDL-SAA)and high density lipoprotein-paraoxonase-1(HDL-PON1)were detected by ELISA.The HDL-SAA/HDL-ApoAI ratio was calculated,and the content of high density lipoprotein 1 phospholipid sphingosinol(HDL-S1 P)was determined by HPLC.Results Serum HDL levels of the model group decreased at week 24(P<0.05),which were significantly lower than those of the normal group at week 24(P<0.05).The levels of HDL-ApoAI,HDL-PON1 and HDL-S1 p in serum of the pigs in the model group at week 24 were significantly lower than those both in the model group at week 0 and the normal group in the same period(P<0.01).Meanwhile,serum HDL-SAA level and HDL-SAA/HDL-ApoAI ratio at week 24 in the model group were significantly increased(P<0.01).Conclusion The serum HDL level of hyperlipidemia pigs with spleen deficiency and phlegm-turbid syndrome was significantly decreased,and the composition of the serum was changed,leading to the decrease of reverse cholesterol transportation,anti-inflammatory and anti-oxidation effects,and the pigs became“defunctional”.
