摘要
目的探讨核转运蛋白α-2(KPNA2)在肝细胞癌中的表达与预后意义及其在肿瘤免疫中的重要作用。方法分别从多平台数据库中下载肝细胞癌数据,通过R语言进行数据整理和筛选,用Wilcoxon-test进行表达差异分析,Kruskal-test和Cox回归模型进行临床差异与预后分析,最后用ssGSEA和Cibersot算法进行免疫细胞浸润与免疫功能相关研究。结果KPNA2在11个肝细胞癌样本组织中的表达水平明显高于癌旁组织(P<0.01),其高表达与肝细胞癌患者不良预后及肿瘤分期等密切相关[风险比(HR)>1,P<0.05)。KPNA2还可影响肝细胞癌中的抗原提呈与白细胞介素反应等免疫功能(P<0.01),并与包括B细胞,CD4^(+)T细胞等在内的10种免疫细胞的浸润水平明显相关(P<0.05)。结论KPNA2可促进肝细胞癌发生发展,并能作为肝细胞癌的独立预后因子,同时其通过调节免疫细胞浸润可影响肿瘤微环境。
Objective To investigate the expression and prognostic significance of karyopherin alpha 2(KPNA2)in hepatocellular carcinoma(HCC)and its important role in tumor immunity.Methods Hepatocellular carcinoma data were downloaded from multi-platform databases,sorted and screened by R language,expression differences were analyzed by Wilcoxon-test,clinical differences and prognosis analysis were evaluated by Kruskal-test and Cox regression analyses.Immune-related studies were conducted by ssGSEA and Cibersot algorithms.Results The expression level of KPNA2 in 11 HCC samples was significantly higher than that in adjacent tissues(P<0.01),and its high expression was closely related to poor prognosis and tumor stages of HCC patients[hazard ratio(HR)>1,P<0.05].KPNA2 also affected the immune function of hepatocellular carcinoma,such as antigen presentation and interleukin reaction(P<0.01),and was significantly correlated with the infiltration level of 10 kinds of immune cells,including B cells,CD4^(+)T cells(P<0.05).Conclusion KPNA2 can promote the progress of hepatocellular carcinoma and act as an independent prognostic factor for hepatocellular carcinoma.Meanwhile,KPNA2 can affect tumor microenvironment by regulating immune cell infiltration.
