胃癌组织中EPAS1的表达及临床意义

Expression of EPAS1 in gastric cancer tissue and its clinical significance

目的探讨EPAS1表达与胃癌临床病理特征的关系及临床意义。方法TCGA数据库分析胃癌和正常组织中EPAS1的表达情况及与预后的关系。收集95例胃癌标本,采用免疫组化法检测EPAS1在胃癌、癌旁、腺瘤组织中的表达,并分析EPAS1表达与胃癌临床病理特征和预后的关系。使用qRT-PCR和Western blot法检测EPAS1在胃癌细胞系中的表达情况。结果胃癌组织中EPAS1高表达58例,EPAS1低表达37例。胃癌组织中EPAS1免疫组化表达得分显著高于癌旁、腺瘤组织(t=6.213、10.691,P<0.05),在转录和翻译水平EPAS1在胃癌细胞系中均显著高表达,EPAS1高表达组患者的总生存期低于EPAS1低表达组(t=7.149,P<0.05)。EPAS1高表达组的原发灶深度浸润和低分化程度均高于低表达组(t=3.138、2.275,P<0.05)。Cox多因素分析结果显示:EPAS1表达(HR:2.095,95%CI:1.019~4.307,P<0.001)是总生存期的独立预测因子。结论EPAS1在胃癌组织中高表达,与原发灶浸润和分化程度显著相关,可能是胃癌预后的标志物。

Purpose To investigate the expression of EPAS1 in gastric cancer(GC)tissue and its correlation with clinicopathological characteristics and prognosis.Methods Using TCGA database we analyzed the expression of EPAS1 in gastric cancer tissue and normal tissue and its relationship with prognosis.The expression of EPAS1 in GC tissue,paracancerous tissue and adenoma tissue was detected by immunohistochemical method.The expression of EPAS1 in gastric cancer cell lines was detected by qRT-PCR and Western blot.A total of 95 patients with postoperative pathologically confirmed GC were enrolled,and the correlation of EPAS1 expression with clinicopathological features and prognosis was analyzed.Results There were 58 cases in EPAS1 high expression group and 37 cases in EPAS1 low expression group in GC tissue.The immunohistochemical expression score of EPAS1 in GC tissues was significantly higher than that in adjacent tissues and adenoma tissues(t=6.213,10.691,P<0.05).EPAS1 was significantly overexpressed in gastric cancer cell lines at both transcriptional and translational levels.The overall survival(OS)of the EPAS1 high expression group was significantly lower than that of the EPAS1 low expression group(t=7.149,P<0.05).The deep infiltration and low differentiation degree of the primary tumor in the high expression group were significantly higher than those in the low expression group(t=3.138,2.275,P<0.05).Multivariate Cox analysis showed that EPAS1 expression(HR:2.095,95%CI:1.019-4.307,P<0.001)was an independent predictor of OS.Conclusion EPAS1 is highly expressed in gastric cancer tissue and is significantly correlated with the degree of invasion and differentiation of the primary tumor.It may be a prognostic marker for gastric cancer patients.
