摘要
研究福建省龙岩市象洞鸡养殖群体的遗传多样性及起源进化关系,为其选育和遗传改良提供理论依据。本研究通过PCR扩增与测序技术获得象洞鸡细胞色素b(Cytb)基因和线粒体控制区(D-loop)部分序列,采用DNAMAN软件进行序列拼接,Dna SP和Mega软件分析序列变异和种群的遗传多样性。结果显示:获得象洞鸡Cytb序列为810 bp,检测到7个核苷酸多态位点,定义了6种单倍型,核苷酸多样度为0.00199,单倍型多样度为0.713;获得D-loop序列为565 bp,检测到25个核苷酸多态位点,定义了16种单倍型,核苷酸多样度为0.001173,单倍型多样度为0.940。系统发育树分析显示象洞鸡有4个母系来源。该研究结果从分子水平为象洞鸡遗传资源保护和开发利用提供了理论参考。
To study the genetic diversity and evolution relationship of breeding Xiangdong chicken in Longyan,Fujian province,in order to provide a theoretical basis for the breeding and genetic improvement.In this study,the Cytb gene and D-loop partial sequences were obtained by PCR amplification and sequencing technology,and the sequence variation and genetic diversity of the population were analyzed by DNAMAN,Dna SP and Mega software.The results showed that the Cytb sequence was 810 bp,7 nucleotide polymorphic sites were detected,and 6 haplotypes were defined.The nucleotide diversity was 0.00199,and the haplotype diversity was 0.713.The D-loop sequence was565 bp,25 nucleotide polymorphic sites were detected,and 16 haplotypes were defined.The nucleotide diversity was 0.001173,and the haplotype diversity was 0.940.Phylogenetic tree analysis showed that there may be more than 4 maternal origins in Xiangdong chicken population.The results could provided a theoretical reference for the conservation and utilization of genetic resources of Xiangdong chicken from the molecular level.
