富硒青钱柳、黄精单方及复方对D-半乳糖致衰老小鼠抗氧化作用的研究 被引量:1

摘要 为了研究富硒青钱柳、黄精单方及富硒青钱柳黄精复方对D-半乳糖致衰老小鼠模型的抗氧化作用,将昆明小鼠随机分为对照组、模型组、维生素E组、富硒青钱柳组、黄精组、富硒青钱柳黄精复方低剂量组和富硒青钱柳黄精复方高剂量组,连续灌胃42 d后摘眼球取血,测定各组小鼠肝血清以及脑、肝脏匀浆中的谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)活性及丙二醛(malondialdehyde,MDA)含量,采用HE染色观察脑组织、肝组织病理学改变,结果显示,与对照组相比,模型组小鼠血清、全脑及肝脏匀浆中SOD、CAT和GSH-Px的水平明显下降,而MDA的水平显著上升(P<0.05或P<0.01)。与模型组相比,富硒青钱柳组小鼠显著提高脑组织GSH-Px、血清和肝脏SOD、血清CAT的活性(P<0.05或P<0.01),显著降低脑组织中MDA的含量;黄精组小鼠显著提高脑组织GSH-Px、血清和肝脏SOD、血清和脑组织CAT的活性(P<0.05或P<0.01);富硒青钱柳黄精复方组小鼠显著提高血清以脑、肝脏匀浆中的SOD、CAT及GSH-Px的活性(P<0.05或P<0.01),显著降低脑组织中MDA的含量(P<0.05或P<0.01)。HE染色发现富硒青钱柳黄精复方对衰老小鼠脑组织和肝组织具有保护作用。实验证明,富硒青钱柳黄精复方较单方能有效提高D-半乳糖致衰老小鼠模型抗氧化能力,达到延缓衰老的作用。 To study the antioxidant effects of Selenium-rich Cyclocarya paliurus and Polygonatum sibiricum single, Selenium-rich Cyclocarya paliurus and Polygonatum sibiricum compound on D-galactose induced aging mice. Kunming mice were randomly divided into the control group, model group, vitamin E group, Selenium-rich Cyclocarya paliuruss group, Polygonatum sibiricum group, Selenium-rich Cyclocarya paliurus and Polygonatum sibiricum compound low-dose group and compound high-dose group. The activities of glutathione peroxidase(GSH-Px), superoxide dismutase(SOD), catalase(CAT) and malondialdehyde(MDA) in serum, brain and liver of each group were determined. Histopathological changes of brain and liver tissues were observed by HE staining. The results showed that, compared with the control group, SOD, CAT and GSH-Px levels in serum, brain and liver of model group were significantly decreased, while MDA levels were significantly increased(P < 0.05, or P < 0.01). Compared with the model group, the activities of GSH-Px in brain tissue, SOD in serum and liver and CAT in serum of Selenium-enriched Cyclocarya paliurus group were significantly increased(P < 0.05, or P < 0.01), MDA content in brain significantly decreased;The activities of GSH-PX in brain tissue, SOD in serum and liver, CAT in serum and brain tissue in Polygonatum sibiricum group were significantly increased(P < 0.05, or P < 0.01);The activities of SOD, CAT and GSH-Px in serum homogenate of brain and liver in Selenium-rich Cyclocarya paliurus and Polygonatum sibiricum compound groups were significantly increased(P < 0.05, or P < 0.01), significantly decreased MDA content in brain tissue(P < 0.05, or P < 0.01). HE staining showed that Selenium-rich Cyclocarya paliurus and Polygonatum sibiricum compound had protective effects on brain and liver tissues in aging mice. In conclusion, Selenium-rich Cyclocarya paliurus and Polygonatum sibiricum compound can effectively improve the antioxidant capacity of D-galactose induced aging mouse model, and achieve the effect of delaying aging.
