摘要
目的探究正天丸(Zhengtian pills,ZTP)对硝酸甘油诱导的偏头痛大鼠痛觉过敏和畏光的缓解作用及其机制。方法大鼠随机分成对照组、偏头痛模型组和ZTP治疗组,每组15只。用von Frey测痛仪检测大鼠机械痛阈值,评估机械痛觉敏感性;用热水浴法检测大鼠撤尾潜伏期,评估热痛觉敏感性;用黑白箱实验检测观察大鼠的畏光行为;用HE染色观察三叉神经节中的卫星胶质细胞;用免疫荧光染色观察三叉神经节中CGRP和c-Fos的表达。结果偏头痛模型组大鼠机械痛阈值降低,热痛觉阈值降低,表现出明显的痛觉过敏;ZTP可缓解偏头痛大鼠的痛觉过敏现象。黑白箱实验结果显示,偏头痛模型组大鼠待在白箱中的时间明显降低,表现出畏光行为;ZTP可改善偏头痛大鼠的畏光行为。HE染色显示,偏头痛模型组大鼠的三叉神经节中卫星细胞明显增多;ZTP可降低偏头痛大鼠三叉神经节中卫星细胞数。免疫荧光染色表明,偏头痛模型组大鼠三叉神经节中CGRP和c-Fos表达均增加;ZTP可降低偏头痛大鼠CGRP和c-Fos的表达。结论ZTP能缓解硝酸甘油诱导的偏头痛大鼠的痛觉过敏和畏光行为,且这种作用可能与降低中枢敏化相关。
Objective To investigate whether Zhengtian pills(ZTP)have an alleviative effect on hyperalgesia and photophobia in nitroglycerin-induced migraine rats and explore the mechanism.Methods Rats were randomly divided into the control group,migraine model group and ZTP treatment group,with 15 rats in each group.The mechanical pain threshold of rats was measured using the von Frey pain meter to assess the mechanical pain sensitivity.The latent period of rats’withdrawal was measured by the hot water bath method to evaluate the thermal pain sensitivity.Light/dark box testing was applied to observe the photophobia behavior of rats.Satellite cells in trigeminal ganglion were observed by HE staining.The expression of CGRP and c-Fos in trigeminal ganglion was observed by fluorescence staining.Results The mechanical and thermal pain thresholds were decreased in the migraine model group,showing significant hyperalgesia.The light/dark box testing showed rats spent significantly less time in the light box in the migraine model group,showing photophobia behavior.HE staining and immunofluorescence staining showed the number of satellite cells in the trigeminal ganglia of migraine rats were significantly increased,and the expression levels of CGRP and c-Fos in the ganglia were also increased.ZTP could alleviate all the above-mentioned migrainous changes.Conclusion ZTP can alleviate hyperalgesia and photophobia in nitroglycerin-induced migraine rats,and this effect may be associated with the decreased central sensitization.
