摘要
目的观察调胃消痞方对功能性消化不良大鼠胃肠运动及血清干细胞生长因子(SCF)、白细胞介素-6(IL-6)水平的影响,探讨调胃消痞方治疗功能性消化不良的作用机制。方法将40只SD大鼠随机分为空白组、模型组、莫沙必利组、调胃消痞方组,每组10只。除空白组外,其余组大鼠采用夹尾刺激联合不规则喂食7 d的方法建立功能性消化不良模型。造模结束后,调胃消痞方组给予5.00 g/mL调胃消痞方药液灌胃,莫沙必利组给予0.50 mg/mL枸橼酸莫沙必利片剂药液灌胃,空白组与模型组均给予等体积生理盐水灌胃,均每日1次,连续灌胃14 d。比较各组大鼠体重、胃内残留率、小肠推进率,HE染色观察各组大鼠胃窦部组织病理学形态,ELISA法检测各组大鼠血清SCF和IL-6水平。结果与空白组比较,模型组大鼠体重、小肠推进率及血清SCF水平均明显降低(P均<0.05),胃内残留率及血清IL-6水平均明显升高(P均<0.05),大鼠胃窦黏膜腺体中可见少量炎性细胞浸润;与模型组比较,莫沙必利组、调胃消痞方组大鼠体重、小肠推进率及血清SCF水平均明显升高(P均<0.05),胃内残留率及血清IL-6水平均明显降低(P均<0.05),大鼠胃窦黏膜腺体中未见明显炎性细胞浸润;调胃消痞方组大鼠小肠推进率明显高于莫沙必利组(P<0.05),血清IL-6水平明显低于莫沙必利组(P<0.05),体重、胃内残留率、血清SCF水平与莫沙必利组比较差异均无统计学意义(P均>0.05)。结论调胃消痞方可通过升高血清SCF水平和降低IL-6水平,调节功能性消化不良大鼠胃肠运动及减轻炎症反应。
Objective It is to observe the effect of Tiaowei Xiaopi decoction on gastrointestinal motility and the levels of stem cell factor(SCF)and interleukin-6(IL-6)in serum of rats with functional dyspepsia(FD),to explore the mechanism of this decoction in the treatment of FD.Methods Forty SD rats were randomly divided into blank group,model group,mosapri group and Tiaowei Xiaopi decoction group,with 10 rats in each group.The rats in every group except for the blank group were given tail clip stimulation combined with irregular feeding to establish models of FD.After the completion of modeling,the rats in the Tiaowei Xiaopi decoction group were given 5.00 g/mL Tiaowei Xiaopi decoction solution,the rats in the mosapride group were given 0.50 mg/mL mosapride citrate tablet solution,and the rats in the blank group and model group were given the same volume of normal saline,all once per day,continuously treated for 14 days.The body weight,gastric residual rate and intestinal propulsion rate of the rats in each group were compared,the histopathological morphology of gastric antrum of the rats in each group was observed by HE staining,the levels of serum SCF and IL-6 of the rats in each group were detected by ELISA.Results Compared with the blank group,the body weight,intestinal propulsion rate and level of serum SCF of the rats in the model group were significantly decreased(all P<0.05),and the gastric residual rate and level of serum IL-6 were significantly increased(all P<0.05),the infiltration of a few inflammatory cells were found in gastric antrum mucosa glands.Compared with the model group,the body weight,intestinal propulsion rate and level of serum SCF of the rats in the mosapri group and Tiaowei Xiaopi decoction group were significantly increased(all P<0.05),and the gastric residual rate and level of serum IL-6 were significantly decreased(all P<0.05),no obvious infiltration of inflammatory cells were found in gastric antrum mucosa glands.The intestinal propulsion rate of the rats in the Tiaowei Xiaopi decoction group was significantly higher than that in the mosapride group(P<0.05),the serum IL-6 level was significantly lower than that of mosapride group(P<0.05),and there was no significant difference in body weight,gastric residual rate,and serum SCF level compared with mosapride group(P>0.05).Conclusion Tiaowei Xiaopi decoction can regulate gastrointestinal motility and relieve inflammatory response in the rats with FD by increasing serum SCF and decreasing serum IL-6.
