摘要 目的 通过体外实验研究FGFR4在宫颈癌细胞中的表达及其影响宫颈癌发生发展的作用机制。方法 应用实时定量PCR和蛋白质免疫印迹实验检测宫颈癌细胞中FGFR4的mRNA及蛋白表达水平;运用流式细胞术检测FGFR4对宫颈癌细胞周期的影响;CCK-8实验验证FGFR4对宫颈癌细胞增殖的影响;应用Transwell小室实验检测FGFR4对宫颈癌细胞侵袭迁移能力的影响;采用siRNA技术沉默FGFR4基因后观察细胞的耐药性变化,并进行分析。结果 体外实验结果表明,FGFR4在宫颈癌Hela细胞中表达升高(t=20.870,P<0.05),沉默FGFR4基因,宫颈癌Hela细胞生长增殖率降低(t=-13.830,P<0.05),凋亡细胞数增多(t=8.740,P<0.05),NF-κB信号通路蛋白NF-κB p65的表达水平明显降低(t=-5.443,P<0.05),迁移侵袭能力下降(t=-6.421,P<0.05);与单纯顺铂(DDP)组相比,FGFR4-siRNA+DDP组细胞凋亡数显著增加(t=10.220,P<0.05)。结论 FGFR4可能通过促进Hela细胞增殖、侵袭迁移、增强其对顺铂的耐药性以及抗细胞凋亡等途径参与宫颈癌的发生发展。 Objective To study the expression of FGFR4 in cervical cancer cells and the mechanism of affecting the occurrence and development of cervical cancer through in vitro experiments.Methods Real-time quantitative PCR and Western blotting were used to detect the expression of FGFR4 in cervical cancer cells at mRNA and protein levels.Flow cytometry was used to study the effect of FGFR4 on the cycle of cervical cancer cells.CCK-8 experiment verified the effect of FGFR4 on the apoptosis of cervical cancer cells.Transwell chamber experiment was used to detect the influence of FGFR4 on the invasion and migration of cervical cancer cells.The siRNA technique was used to silence the FGFR4 gene, so as to observe and analyze the changes of drug resistance in cells.Results The results of in vitro experiments showed that the expression of FGFR4 increased in cervical cancer Hela cells(t=20.870,P<0.05),and the silencing of the FGFR4 gene decreased the growth and proliferation rate of cervical cancer Hela cells(t=-13.830,P<0.05).The number of apoptosis cells increased(t=8.740,P<0.05),the expression level of NF-κB signaling pathway protein, NF-κB p65 significantly decreased(t=-5.443,P<0.05),and the ability of migration and invasion decreased(t=-6.421,P<0.05).Compared with the pure cis-diamminedichloro-platinum(DDP) group, the number of apoptosis cells in the FGFR4-siRNA+DDP group was significantly increased(t=10.220,P<0.05).Conclusion FGFR4 may participate in the occurrence and development of cervical cancer by promoting cell proliferation, invasion and migration, enhancing drug resistance of cis-diamminedichloro-platinum and resisting apoptosis in Hela cells.
