摘要
支气管哮喘(哮喘)的病理改变包括气道炎症、气道高反应性、气道重塑, 而气道炎症会加重气道重塑与气道高反应性。热休克反应在炎症和纤维化的发生、发展中起到重要作用。热休克蛋白90是热休克蛋白家族成员之一, 可以辅助客户蛋白正确折叠, 调节白细胞介素的释放、肿瘤坏死因子的产生, 上皮-间充质间转化, 成纤维细胞的激活等。其在机体发挥调控作用受到多种机制的调节:翻译后修饰、辅助伴侣因子以及ATP酶的结合。哮喘气道炎症和重塑过程受众多因素的影响, 其中许多因素与HSP90相关。本综述将总结HSP90的翻译后修饰以及与其协同的共伴侣蛋白特异性结合对哮喘气道炎症和气道重塑的影响, 为未来哮喘治疗方式提供新途径和新依据。
Key pathophysiologic features of bronchial asthma(asthma)include airway inflammation,airway hyperresponsiveness,and airway remodeling.Airway sustaining inflammation will increase airway hyperresponsiveness and remodeling.Heat shock response plays an important role in the development of inflammation and fibrosis.Heat shock protein 90(HSP90),one member of heat shock family,helps nascent polypeptides fold in right conformation,mediates cytokines release like interleukin,mediates tumor necrosis factors production,adjusts epithelial mesenchymal transformation,and activates fibroblast.The biological function of HSP is regulated by many mechanisms:post-translational modifications,co-chaperones,and ATPase bindings.Airway inflammation and remodeling are affected by many factors,most of which are related to HSP90.This review will summarize the function of HSP90 post-translational modification and the effects of the specific binding with their co-chaperones on airway inflammation and remodeling,so as to provide a novel method and basis for the treatment of asthma in the future.
