荧光化试剂衍生联合高效液相色谱法测定鱼罐头中组胺含量与污染水平初步分析

Determination of Histamine in Canned Fish by Reagent Derivatization Combined with High Performance Liquid Chromatography

目的:建立荧光化试剂衍生-高效液相色谱法测定7类鱼罐头中组胺的含量。方法:样品采用5%三氯乙酸提取,经除脂、萃取后,利用强荧光试剂丹磺酰氯进行衍生,0.22μm滤膜过滤后,高效液相色谱-紫外法测定。结果:该方法线性相关系数r=0.9999,检出限为10.8mg/kg。所检测的7类30种鱼罐头样品检出率为36.7%,检出的鱼罐头样品中组胺含量均低于300mg/kg,符合国家标准。结论:该方法线性关系良好,检出限较低,测定结果准确,适合用于鱼罐头中组胺含量的检测。

Objective:To establish a reagent derivatization HPLC method for the determination of histamine in 7 categories of canned fish.Methods:The sample was extracted with 5% trichloroacetic acid.After degreasing and extraction, it was derived with dansulfonyl chloride.After 0.22μm membrane filtration, it was determined by HPLC-UV.Results:The linear correlation coefficient of the method was r=0.9999 and the detection limit was 10.8 mg/kg.The detection rate of 7 categories and 30 species of canned fish samples was 36.7%.The content of histamine in canned fish samples was less than 300 mg/kg, which met the national standard.Conclusion:The method has good linear relationship, low detection limit and accurate results.It is suitable for the determination of histamine in canned fish.
