3株枫香优株组培快繁体系的建立 被引量:2

摘要 以枫香优良单株HA-1、HA-2、HA-3的带芽茎段为材料,以WPM+3.0%蔗糖+0.7%琼脂为基本培养基,通过添加不同激素组合进行组织培养研究.结果表明:①初代培养:HA-1适宜的激素配比为1.5 mg/L 6-BA+1.0 mg/L KT+0.05 mg/L NAA+0.1 mg/L IBA;HA-2和HA-3适宜的激素配比均为1.0 mg/L 6-BA+0.05 mg/L NAA.②增殖培养:HA-1适宜的激素组成为2.0 mg/L 6-BA+0.05 mg/L NAA;HA-2适宜的激素组成为1.0 mg/L 6-BA+0.01 mg/L NAA;HA-3适宜的激素组成为1.0 mg/L 6-BA+0.03 mg/L NAA+1.0 mg/L GA_(3).③生根培养:HA-1、HA-2和HA-3生根率最高的激素配比均为1.0 mg/L IBA.生根苗移栽后,三者的成活率均可达到100%.通过建立枫香优株的组培快繁体系,以利于枫香良种的推广. Using the stem segments with buds of the excellent individual plants HA-1,HA-2 and HA-3 of Liquidambar formosana as the material,with WPM+3.0%sucrose+0.7%agar as the basic medium,the tissue culture study was carried out by adding different combinations of hormones.The research shows:①Primary culture:the suitable hormone ratio of HA-1 is 1.5 mg/L 6-BA+1.0 mg/L KT+0.05 mg/L NAA+0.1 mg/L IBA;the suitable hormone ratio of HA-2 and HA-3 is 1.0 mg/L 6-BA+0.05 mg/L NAA.②Proliferation culture:the suitable hormone composition of HA-1 is 2.0 mg/L 6-BA+0.05 mg/L NAA;the suitable hormone composition of HA-2 is 1.0 mg/L 6-BA+0.01 mg/L NAA;the suitable hormone composition of HA-3 is 1.0 mg/L 6-BA+0.03 mg/L NAA+1.0 mg/L GA3.③Rooting culture:the hormone ratio for the highest rooting rate of HA-1,HA-2 and HA-3 is 1.0 mg/L IBA.After the rooted seedlings are transplanted,the survival rate of the three can reach 100%.The establishment of a tissue culture and rapid propagation system of the excellent Liquidambar species is conducive to the popularization of the Liquidambar superior varieties.
