摘要
目的:探讨预服生脉饮加味方对大鼠肾缺血再灌注致急性肾损伤的影响及作用机制。方法:将45只SD大鼠随机分成假手术组、模型组及生脉饮加味方组各15只,3组连续灌胃7d后构建肾缺血再灌注模型,随后分别于造模后第1、3、6天检测大鼠的尿素氮(BUN)、中性粒细胞明胶酶相关脂质运载蛋白(NGAL)mRNA和肾损伤分子-1(KIM-1)以及血肌酐(SCr)的表达、肾小管细胞的凋亡情况及肾组织的病理变化,同时检测Bax蛋白表达、Bcl-2及Caspase-3。结果:与假手术组相比较,第1、3天模型组中肾组织的病理损伤显著,Bcl-2/Bax值降低,差异有统计学意义(P<0.01),血Scr与BUN水平、Caspase-3蛋白表达、肾组织中NGALmRNA、Kim-1表达均升高,差异有统计学意义(P<0.05)。与模型组相比,生脉饮加味方组Caspase-3蛋白表达、Bcl-2/Bax值、凋亡指数,肾组织中Kim-1、NGALmRNA表达均降低,差异有统计学意义(P<0.05),肾组织的病理损伤减轻明显。结论:预服生脉饮加味方对肾脏缺血再灌注所致的急性肾损伤有一定的改善作用,其作用机制可能与上调Bcl-2/Bax值及下调肾组织Caspase-3有关。
Objective Observe the effect of pre-taken modified pulse-activating powder on acute kidney injury induced by renal ischemia-reperfusion in rats and explore its internal mechanism.Methods Forty-five SD rats were randomLy divided into shamoperated group,model group and modified pulse-activating powder group,15 rats in each group,and the renal ischemia-reperfusion model was constructed after 7 days of continuous gavage in the three groups.The expression of urinary BUN,Scr,NGAL mRNA,Kim-1,apoptosis of renal tubular cells and pathological changes of renal tissues were detected on the 1st,3rd and 6th days after modeling,and Bax protein expression,Bcl-2 and Caspase-3 were also detected.Results When compared with the sham-operated group,the pathological damage of renal tissues was significant in the model group on 1st day and 3rd day,and Bcl-2/Bax values were reduced(P<0.01),and blood Scr and BUN levels,Caspase-3 protein expression,and NGAL mRNA and Kim-1 expression in renal tissues were increased(P<0.05).Conclusions Pre-taken modified pulse-activating powder has some ameliorative effect on acute kidney injury caused by renal ischemia-reperfusion,and its mechanism of action may be related to up-regulation of Bcl-2/Bax value and down-regulation of Caspase-3 in renal tissues.
