一测多评法同时测定青皮中5种成分的含量 被引量:2

Simultaneous Determination of the Content of 5 Components in Citri Reticulatae Pericarpium Viride by Quantitative Analysis of Multi-components by Single Marker

摘要 目的:引入一测多评(QAMS)的概念,建立QAMS法以同时测定青皮中5种黄酮类成分的含量。方法:采用UPLC法,乙腈-0.1%甲酸溶液作为流动相,梯度洗脱;检测波长为275 nm;柱温为40℃;进样量为1μL;体积流量为0.3 mL/min。设定内参照物为橙皮苷,计算其与辛弗林、芸香柚皮苷、川陈皮素和橘皮素的相对校正因子,并在多品牌的色谱仪和色谱柱上考察各成分相对校正因子的稳定性和耐用性。结果:辛弗林、芸香柚皮苷、橙皮苷、川陈皮素、橘皮素在各自规定的浓度范围内呈良好线性关系(r>0.9990);5种成分的平均加样回收率分别为101.05%(RSD 2.82%)、96.01%(RSD 2.92%)、96.93%(RSD 1.73%)、94.01%(RSD 2.78%)、103.86%(RSD 2.00%);相对校正因子分别为13.99、1.33、0.76、0.58。一测多评法计算10批青皮中5种成分的含量与外标法测定值间均无明显差异。结论:建立的QAMS法同时测定青皮中5种黄酮类成分的含量简单可行、结果准确,可对青皮药材的验收及质量控制提供参考。 Objective:To introduce the concept of quantitative analysis of multi-components by single marker(QAMS),and establish a QAMS method to simultaneously determine the contents of five flavonoids in citri reticulatae pericarpium viride.Methods:A UPLC method was used,acetonitrile-0.1%formic acid solution was used as mobile phase,gradient elution was performed;the detection wavelength was 275 nm;the column temperature was 40℃;the injection volume was 1μL;the volume flow was 0.3 mL/min.Set the internal standard as hesperidin,calculate its relative correction factor with synephrine,naringenin,tangerine peelin and hesperidin,and investigate the relative correction of each component on multi-brand chromatographs and chromatographic columns factor stability and durability.Results:Synephrine,naringin,hesperidin,chuanmenetin,and citrus peretin showed a good linear relationship(r>0.9990)in their respective specified concentration ranges;the average sample recovery rates of the five components were 101.05%(RSD 2.82%),96.01%(RSD 2.92%),96.93%(RSD 1.73%),94.01%(RSD 2.78%),103.86%(RSD 2.00%);the relative correction factors were 13.99,1.33,0.76,0.58,respectively.There was no significant difference between the content of 5 components in 10 batches of citri reticulatae pericarpium viride calculated by QAMS method.Conclusion:The established QAMS method is simple and feasible,and the results are accurate,which can provide reference for the acceptance and quality control of citri reticulatae pericarpium viride medicinal materials.
