辣椒素改善实验小鼠急性肺损伤氧化应激的研究 被引量:2

摘要 目的:探讨辣椒素对内毒素所致实验小鼠急性肺损伤氧化应激的改善作用。方法:将白化的家鼠亚系(BALB/c)小鼠120只随机分为4组:对照组(n=30)、辣椒素对照组(n=30)、内毒素组(n=30)、辣椒素+内毒素干预组(n=30),分别于2 h、4 h、6 h时点麻醉后处死,留取肺组织样本。化学比色法检测小鼠丙二醛(MDA)水平变化;一氧化氮(NO)还原酶法测定氮氧化合物(NOX)水平变化;酶联免疫吸附试验法(ELISA)检测小鼠肺组织肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)表达变化,组织切片苏木精-伊红(HE)染色后观察各组小鼠肺组织病理学改变。结果:与对照组比较,内毒素组小鼠肺组织MDA、NOX、TNF-α、IL-1β水平均明显升高(P<0.05),辣椒素对照组则变化不明显(P>0.05),与内毒素组比较,辣椒素+内毒素干预组小鼠肺组织MDA、NOX、TNF-α、IL-1β水平均明显降低(P<0.05)。光镜下,内毒素组4 h、6 h较对照组肺组织损伤明显,辣椒素对照组则无明显变化,辣椒素+内毒素干预组肺损伤程度较内毒素组轻。结论:氧化应激是内毒素血症小鼠急性肺损伤发生的重要环节,辣椒素可通过降低SOD和NOX水平,并通过降低TNF-α和IL-1β表达水平从而减轻急性肺损伤小鼠氧化应激损伤。 Objective:To study the effect of capsaicin on the oxidative stress in acute lung injury induced by endotoxin in mice.Methods:A total of 120 BALB/c mice were randomized into control group(n=30),capsaicin group(n=30),endotoxin group(acute lung injury group)(n=30),and capsaicin+endotoxin group(n=30).They were killed at 2,4 and 6 h after anesthesia,respectively,and lung tissue samples were collected.The level of malondialdehyde(MDA)was determined by chemical colorimetry and level of nitrogen oxides(NOX)by nitric oxide(NO)reductase method.Levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in lung tissues of mice were detected by enzyme-linked immunosorbent assay(ELISA).Pathological changes of lung tissue were observed based on hematoxylin and eosin(HE)staining.Results:The levels of MDA,NOx,TNF-α,and IL-1βin the lung tissue of mice in the endotoxin group were increased compared with those in the control group(P<0.05),but the levels in the capsaicin group showed no significant changes compared with those in the control group(P>0.05).Levels of MDA,NOx,TNF-α,and IL-1βin the lung tissue of mice in the capsaicin+endotoxin group were lower than those in the endotoxin group(P<0.05).Under light microscope,the lung tissue damage in endotoxin group was more obvious than that in control group at 4 h and 6 h,and capsaicin group showed no significant variation.In addition,the damage in the capsaicin+endotoxin group was milder than that in endotoxin group.Conclusion:Oxidative stress is an important link in the development of acute lung injury in endotoxemia mice.Capsaicin can alleviate oxidative stress injury in acute lung injury mice by reducing levels of superoxide dismutase(SOD),NOX,TNF-α,and IL-1β.
