奶牛乳房炎肠杆菌科主要病原菌OmpA理化特性、同源性及抗原表位生物信息学分析 被引量:1

摘要 奶牛乳房炎(Bovine Mastitis)是严重影响奶牛养殖业和奶业健康发展最为重要的疾病,以埃希氏大肠杆菌(Escherichia coli)为代表的革兰氏阴性肠杆菌科细菌是引起该病的重要病原菌。外膜蛋白A(OmpA)是肠杆菌科细菌外膜上的重要成分。研究表明,E.coli的OmpA具有良好的免疫原性,是预防E.coli引起奶牛乳房炎的重要候选疫苗抗原。但对引起奶牛乳房炎的肠杆菌科细菌OmpA的同源性及免疫原性分析缺乏报道。研究选取报导的引起奶牛乳房炎的牛源E.coli、克雷伯氏菌、沙门氏菌、福氏志贺氏菌、粘质沙雷氏菌、阴沟肠杆菌、弗氏柠檬酸菌和奇异变形杆菌共29株,用生物信息学方法分析它们OmpA的理化性质、氨基酸序列同源性、CD4+T细胞表位和线性B细胞表位。结果显示,这些细菌OmpA分别由346~359个氨基酸组成,无规则卷曲占比最大,稳定性好且均为亲水性,抗原性良好;与E.coli的OmpA同源性为97.2%~69.3%;都存在着1个高度同源的共同CD4+T细胞表位和6个同源性各异的共同线性B细胞表位。结果为进一步研究OmpA作为预防肠杆菌科细菌引起奶牛乳房炎候选疫苗和建立血清学检测方法提供参考。 Bovine mastitis was a significant disease that seriously affected the development of dairy farming and dairy industry.Gram-negative bacteria from enterobacteriaceae,for example Escherichia coli,were important pathogens causing bovine mastitis.Outer membrane protein A(OmpA)was an important component on the outer membrane of enterobacteriaceae bacteria.Researches had shown that E.coli OmpA had good immunogenicity and was an important vaccine candidate for the prevention of bovine mastitis induced by E.coli.However,there were few reports on the physiochemical characteristics,homology and immunogenicity of OmpA from E.coli and the enterobacteriaceae bacteria causing mastitis.29 strains of E.coli,Klebsiella pneumoniae,Salmonella from cattle,Shigella flexneri,Serratia marcescens,Enterobacter cloacae,Citrobacter freundii and Proteus mirabilis were selected for bioinformatics analysis on physiochemical characteristics,homology and antigen epitopes of their OmpA.The results showed that the OmpA of these bacteria were composed of 346-359 amino acids,with the largest proportion of irregular curl,good stability and hydrophilicity,showing good antigenicity.The homology of OmpA amino acid sequenced from these bacteria was 97.2%~69.3%compared with that from E.coli.There were one common immunodominant CD4+T cell epitope and six common immunodominant B cell epitopes on each OmpA of these bacteria.This analysis provided an important reference for further developing a vaccine using OmpA as antigen against the mastitis and establishing corresponding detection methods.
