基于TCGA鉴定CCNB2在HCC中的临床意义

Expression and Significance of CCNB2 in Hepatocellular Carcinoma Based on TCGA Databases

背景:肝细胞癌(HCC)进展过程复杂,涉及多个基因发生改变.鉴于数据库和生物学工具的发展,寻找了新的生物标记物用于预测HCC患者预后和诊断的价值.方法:从数据库下载样本数据,统计CCNB2在HCC表达情况.使用Kaplan-Meier生存曲线、ROC曲线、时间依赖生存曲线分析CCNB2在诊断和预后中的价值.结果:CCNB2在HCC中高表达,并且与年龄、T分期、病理分期相关,单-多因素Cox分析得出CCNB2是预后的潜在危险因素.在HCC患者中,Kaplan-Meier生存曲线显示CCNB2高表达组获得较低的总生存期(OS)、疾病特异性生存期(DSS)、无进展间期(PFI).ROC曲线下面积(AUC)能很好地鉴别正常组织和恶性肿瘤.CCNB2表达越高,1年、3年、5年生存率越低.结论:CCNB2在HCC中异常表达且与预后不良相关,可能是潜在靶向治疗新标志物.

Background:The progression of hepatocellular carcinoma(HCC)is complex and involves changes in multiple genes.In view of the development of databases and biological tools,we searched for new biomarkers to predict the value of prognosis and diagnosis of HCC patients.Methods:the sample data were downloaded from the database and the expression of CCNB2 in HCC was analyzed.Kaplan Meier survival curve,ROC curve and time-dependent survival curve were used to analyze the value of CCNB2 in diagnosis and prognosis.Results:CCNB2 was highly expressed in HCC and was significantly correlated with age,T stage and pathological stage.Single-multivariate Cox analysis showed that CCNB2 was a potential risk factor for prognosis.In HCC patients,Kaplan Meier survival curve showed that the CCNB2 high expression group achieved lower overall survival(OS),disease-specific survival(DSS),and pro⁃gression free interval(PFI).The area under ROC curve(AUC)can well distinguish normal tissue from malignant tumor.The higher the expression of CCNB2,the lower the 1-,3-,and 5-year survival rates.Conclusion:CCNB2 is abnormally expressed in HCC and is associated with poor prognosis,which may be a new marker of potential targeted therapy.
