超高效液相色谱-串联质谱法同时测定化妆品植物提取物原料中13种黄酮类化合物 被引量:6

Simultaneous Determination of 13 Flavonoids in Cosmetic Plant Extracts by Ultra Performance Liquid Chromatography Tandem Mass Spectrometry

摘要 目的:建立同时测定常用化妆品植物提取物原料中13种黄酮类化合物超高效液相色谱-串联质谱法,为评估植物提取物作为化妆品原料的安全性和规范植物提取物在化妆品中的应用提供理论依据。方法:采用液质联用法,梯度洗脱分离,质谱采用正离子模式,多反应监测。结果:在相应的浓度范围内,13种黄酮类化合物的回归曲线在各自浓度范围内线性良好,相关系数均大于0.99。在低、中、高3个加标水平的平均回收率为86.2%~109.1%,相对标准偏差为1.8%~6.3%,13种黄酮类化合物检出限为0.5~5.0μg·kg^(-1),定量限为为2.0~20μg·kg^(-1),收集的13批植物提取物中,4批检出黄酮类化合物。结论:该方法准确、分析速度快、操作简便、灵敏度高、能满足化妆品植物提取物原料中13种黄酮类化合物的定性筛查和定量检测要求。 Objective:To establish an ultra high performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS)method for the simultaneous determination of 13 flavonoids in common cosmetic plant extracts,so as to provide a theoretical basis for evaluating the safety of plant extracts as cosmetic raw materials and standardizing the application of plant extracts in cosmetics.Methods:LC-MS method.Gradient elution was used for separation.Positive ion mode was used for mass spectrometry.Multi reaction monitoring was used.Results:In the corresponding concentration range,the regression curves of 13 flavonoids had good linearity in their respective concentration ranges,and the correlation coefficients were all greater than 0.99.The average recoveries at low,medium and high spiking levels were 86.2%~109.1%,the relative standard deviation was 1.8%~6.3%,and the detection limit of 13 flavonoids was 0.5~5.0μg·kg^(-1),LOQ is 2.0~20μg·kg^(-1),among 13 batches of plant extracts collected,flavonoids were detected in 4 batches.Conclusion:The method is accurate,fast,simple and sensitive,and can meet the requirements of qualitative screening and quantitative detection of 13 flavonoids in cosmetic plant extracts.
