色胺酮对小鼠血小板扩展和粘附及斑块回缩的作用

Effects of tryptanthrin on platelet spreading,adhesion,and clot retraction in mice

目的探讨色胺酮对小鼠血小板扩展、粘附及斑块回缩的作用。方法昆明种小鼠麻醉采血后制备小鼠洗涤血小板、并调整数目为5×10^(9)个/L,分为空白组(不做任何处理)、对照组(溶剂)、色胺酮组(10μmol/L色胺酮)及阳性药组(0.5 mg/L替罗非班),采用荧光显微镜观察各组小鼠血小板的扩展和粘附情况,并使用ImageJ软件评估各组小鼠血小板的扩散面积和粘附数目;调整洗涤血小板数目为200×10^(9)个/L,分为空白组(不做任何处理)、对照组(溶剂)、色胺酮低剂量组(5μmol/L色胺酮)、色胺酮高剂量组(10μmol/L色胺酮)及阳性药组(0.5 mg/L替罗非班),采用斑块回缩实验观察各组小鼠血小板斑块回缩的程度。结果在纤维蛋白原表面上进行扩展后,对照组小鼠血小板单个平均扩展面积较空白组增加(P<0.01),色胺酮组和阳性药组小鼠血小板单个平均扩展面积较对照组减少(P<0.05或P<0.01);在胶原表面上粘附后,对照组和阳性药组小鼠血小板的粘附数目较空白组增加(P<0.05),色胺酮组小鼠血小板的粘附数目较对照组减少(P<0.05);与对照组相比,色胺酮低、高剂量组小鼠血小板斑块回缩程度被抑制,阳性药组小鼠血小板未见斑块回缩。结论色胺酮可抑制小鼠血小板扩展、粘附及斑块回缩功能。

Objective To investigate the effects of tryptanthrin on mouse platelet spreading,adhesion and clot retraction.Methods Kunming mice were anesthetized and drawn blood to prepare the rinsed platelet.The number of rinsed platelets were adjust to 5×10^(9)/L,and they were divided into blank group(without any treatment),control group(solvent),tryptanthrin group(10μmol/L tryptanthrin)and positive drug group(0.5 mg/L tirofiban).Fluorescence microscope was used to observe the spreading and adhesion of platelets in each group,and ImageJ software was adopted to evaluate the spreading area and adhesion number of mouse platelets in each group.Rinsed platelets number was adjusted to 200×10^(9)/L and were divided into blank group(without any treatment),control group(solvent),tryptanthrin low dose group(5μmol/L tryptanthrin),tryptanthrin high dose group(10μmol/L tryptanthrin)and positive drug group(0.5 mg/L tirofiban).The clot retraction experiment was used to observe the degree of mouse platelet clot retraction in each group.Results After spreading on fibrinogen surface,average spreading area of mouse platelet in control group increased compared with blank group(P<0.01);average spreading area of mouse platelet in tryptanthrin group and the positive drug group decreased compared with control group(P<0.05 or P<0.01).After adhesion to the surface of collagen,the number of mouse platelet adhesion in control group and positive drug group increased compared with blank group(P<0.05).Compared with control group,the clot retraction of mouse platelets in the tryptanthrin low and high-dose groups was inhibited,and the clot retraction of platelets was not observed in positive drug group.Conclusion Tryptanthrin has the effect of inhibiting mouse platelet spreading,adhesion,and clot retraction.
