摘要
目的:检测TSIX(X inactive specific transcript,antisense)与XIST(X inactive specific transcript)在乳腺癌中的表达,分析两者相关性,并评估其临床病理学价值。方法:利用BCIP数据库分析TSIX与XIST在乳腺癌及正常乳腺组织中的表达差异。收集100例患者乳腺癌组织,应用qRT-PCR技术检测TSIX与XIST的表达,绘制ROC曲线,根据尤登指数找出截点值,进行分组,分析其与临床病理参数之间的关系。结果:数据库发现,相比于正常乳腺组织,乳腺癌组织中TSIX表达较高,而XIST表达较低。通过qRT-PCR技术检测同样发现,乳腺癌组织中TSIX表达升高,而XIST表达降低,且两者呈负相关关系(P<0.01)。根据ROC曲线尤登指数最大值,确定TSIX截点值为1.9059;XIST截点值为0.9928。进一步分析发现,TSIX与淋巴结转移(P=0.003)及TNM分期(P=0.037)呈正相关,而XIST仅与淋巴结转移呈负相关(P=0.006)。不仅如此,TSIX阳性而XIST阴性者与淋巴结转移及较晚的TNM分期密切相关。结论:乳腺癌组织中TSIX高表达,而XIST表达较低,两者呈负相关;TSIX与淋巴结转移及TNM呈正相关,而XIST仅与淋巴结转移相关;且联合检测具有更好的临床病理评估作用。
Objective:To detect the expression of TSIX and XIST in breast cancer,analyze their correlation,and evaluate their clinicopathological value.Methods:Differences of TSIX and XIST expression between breast tumors and normal breast tissues were analyzed by BCIP database.Expressions of TSIX and XIST of 100 breast cancer tissues were detected by qRT-PCR.The ROC curve was drawn,and the cut-off values were identified according to Yuden index.Then,relationships of TSIX and XIST with clinicopathological parameters were analyzed.Results:Compared to normal breast tissues,the database found higher expression of TSIX and lower expression of XIST in breast tumors.Through qRT-PCR,expression of TSIX was increased while XIST was decreased in breast tumors,with a negative correlation between them(P<0.01).According to the maximum value of Yuden index of ROC curve,the TSIX cut-off value was 1.9059 and XIST cut-off value was 0.9928.Further analysis showed that TSIX was positively correlated with lymph node metastasis(P=0.003)and TNM stage(P=0.037),but XIST was only negatively correlated with lymph node metastasis(P=0.006).Moreover,patients with positive TSIX and negative XIST were strongly associated with lymph node metastasis and later TNM staging.Conclusion:The expression of TSIX is higher in breast tumors,but XIST is lower,in comparison of normal tissues.In addition,TSIX and XIST have a strongly negative correlation.TSIX is positively correlated with lymph node metastasis and TNM,while XIST is only correlated with lymph node metastasis.And the combined detection of TSIX and XIST might be a better method of clinicopathological evaluation.
