SOX11、miR-15b-5p在子宫内膜癌组织中表达及与患者临床病理特征的关系

The expression of SOX11 and miR-15b-5p in endometrial carcinoma and their relationship with the clinicopathological characteristics of patients

目的 探讨转录因子性别决定区Y框蛋白11(SOX11)、微小RNA-15b-5p(miR-15b-5p)在子宫内膜癌组织中表达及与患者临床病理特征的关系。方法 选取2015年10月至2016年10月120例惠州市第一人民医院行手术治疗的子宫内膜癌患者癌组织及癌旁组织标本。采用免疫组化法检测组织中SOX11的表达,采用实时荧光定量PCR(qRT-PCR)法检测组织中SOX11mRNA、miR-15b-5p的表达水平;Pearson法分析子宫内膜癌组织中SOX11mRNA与miR-15b-5p表达水平相关性;Kaplan-Meier法分析SOX11和miR-15b-5p表达水平与子宫内膜癌患者5年生存率的关系;Cox回归分析影响子宫内膜癌患者总生存的危险因素。结果 子宫内膜癌组织中miR-15b-5p表达水平低于癌旁组织,SOX11蛋白阳性率、SOX11 mRNA表达水平高于癌旁组织(P<0.05)。SOX11蛋白及miR-15b-5p表达与患者淋巴结转移、FIGO分期、肌层浸润有关(P<0.05)。miR-15b-5p在SOX11的3’UTR存在相应的结合位点,子宫内膜癌组织中SOX11mRNA和miR-15b-5p呈负相关(P<0.05)。子宫内膜癌组织中miR-15b-5p高表达、SOX11阴性患者5年生存率明显高于miR-15b-5p低表达、SOX11阳性患者(P<0.05)。FIGO分期Ⅲ期、淋巴结转移、肌层浸润>1/2、miR-15b-5p低表达、SOX11蛋白阳性表达是影响子宫内膜癌患者总生存的独立危险因素(P<0.05)。结论 SOX11蛋白及mRNA在子宫内膜癌组织中呈高表达,miR-15b-5p在子宫内膜癌组织中呈低表达,二者表达水平呈负相关,与患者高FIGO分期、淋巴结转移、肌层浸润增加及5年生存率降低密切相关,有望成为预测子宫内膜癌患者预后的有效指标。

Objective To investigate the expression of SRY-related HMG box 11(SOX11) and microRNA-15b-5p(miR-15b-5p) in endometrial cancer tissues and their relationship with the clinicopathological characteristics of patients.Methods The specimens of cancer tissues and adjacent tissues of 120 patients with endometrial cancer who underwent surgery in Huizhou First Hospital from October 2015 to October 2016 were selected. The expression of SOX11 in tissues was detected by immunohistochemistry, and the expression levels of SOX11 mRNA and miR-15b-5p in tissues was detected by real time fluorescent quantitative PCR(qRT-PCR). Pearson method was used to analyze the correlation between SOX11 mRNA and miR-15b-5p expression in endometrial carcinoma. Kaplan-Meier method was used to analyze the relationship between the expression levels of SOX11 and miR-15b-5p and the 5-year survival rate of patients with endometrial cancer. Cox regression analysis was used to analyze the risk factors affecting the overall survival of patients with endometrial cancer. Results The expression level of miR-15b-5p in endometrial carcinoma was lower than that in adjacent tissues, and the positive rate of SOX11 protein and the expression level of SOX11 mRNA was higher than that in adjacent tissues(P<0.05). The expression of SOX11 protein and miR-15b-5p were related to lymph node metastasis, FIGO stage and myometrial infiltration(P<0.05).miR-15b-5p had corresponding binding sites in the 3’UTR of SOX11, there was a negative correlation between SOX11 mRNA and miR-15b-5p in endometrial carcinoma tissues(P<0.05). The 5-year survival rate of patients with high expression of miR-15b-5p and negative SOX11 in endometrial carcinoma was significantly higher than that of patients with low expression of miR-15b-5p and positive SOX11(P<0.05). FIGO stage Ⅲ, lymph node metastasis, myometrial invasion >1/2, low expression of miR-15b-5p and positive expression of SOX11 protein were independent risk factors affecting the overall survival of patients with endometrial cancer(P<0.05). Conclusion SOX11 protein and mRNA were highly expressed in endometrial carcinoma and miR-15b-5p was low expressed in endometrial carcinoma. Their expression levels were negatively correlated with high FIGO stage, lymph node metastasis, increased myometrial invasion and reduced 5-year survival rate. It is expected to become an effective index to predict the prognosis of patients with endometrial carcinoma.
