氢气干预改善动脉粥样硬化易感小鼠血浆脂蛋白相关磷脂酶A2活力及溶血磷脂酰胆碱水平

摘要 目的探究氢气干预在动脉粥样硬化早期对脂蛋白相关酶和脂质代谢的影响。方法低密度脂蛋白受体基因敲除(LDLr-/-)小鼠高脂饲料饲喂4周,构建动脉粥样硬化易感小鼠模型,同时饮用富氢水及4%氢气吸入联合干预,检测小鼠血脂情况、疾病标志物水平、血浆中多种溶血磷脂酰胆碱(lysophosphatidylcholine,LPC)分子水平及脂蛋白相关磷脂酶A2(lipoprotein⁃related phospholipase A2,Lp⁃PLA2)活力。结果氢气干预可以降低血浆甘油三酯水平及肝脏损伤标志物水平,降低血浆中多种LPC分子水平,改善Lp⁃PLA2活力。结论氢气干预可在动脉粥样硬化易感小鼠中发挥作用,可能通过调节Lp⁃PLA2活性及降低LPC水平缓解动脉粥样硬化的进展。 Objective:To explore the effect of hydrogen intervention on lipoprotein-related enzyme and lipid metabolism in the early stage of atherosclerosis.Methods:Low density lipoprotein receptor-deficient(LDLR-/-)mice were fed with a high-fat diet for four weeks to construct anatherosclerosis-susceptible mice model.Meanwhile,the mice were intervened with a combination of hydrogen-rich water drinking and 4%hydrogen inhalation.After the intervention,the levels of plasma lipids,disease biomarkers,lysophosphatidylcholine(LPC)species,and lipoprotein-related phospholipase A2(Lp⁃PLA2)activity were measured.Results:the levels of plasma triglycerides and liver injury markers were reduced,most of the LPC species were decreased,and the activity of LP-PLA2 was decreased after hydrogen intervention.Conclusion:Hydrogen intervention can play a role in atherosclerosis-susceptible mice,possibly by decreasing LPC levels through regulating the activity of Lp-PLA2.
