植物甾醇纳米粒降低高胆固醇HepG2细胞内胆固醇的作用效果研究 被引量:1

Effect of Phytosterol Nanoparticles on Reducing Intracellular Cholesterol in Hypercholesterolemic HepG2 Cells

摘要 本文通过胆固醇对人肝癌细胞株HepG2细胞进行诱导,构建高胆固醇HepG2细胞模型,再用不同浓度的植物甾醇纳米粒进行干预,测定总胆固醇、总胆汁酸和抗氧化指标,探究植物甾醇纳米粒的降胆固醇效果。结果表明,植物甾醇纳米粒不影响细胞活力,无细胞毒性,能有效降低高胆固醇细胞内胆固醇含量以及提高细胞内抗氧化指标超氧化物歧化酶(Superoxide Dismutase,SOD)和过氧化氢酶(Catalase,CAT)的活力,且存在显著的剂量效应,对培养基中胆汁酸含量没有显著影响。说明植物甾醇纳米粒具有良好的降胆固醇效果,同时能提高细胞抗氧化活性。 In this study,the human hepatoma cell line HepG2 was induced by cholesterol to construct a high cholesterol HepG2 cell model,and then different concentrations of phytosterol nanoparticles were used to intervene,to determine total cholesterol,total bile acid and antioxidant indicators,and to explore the cholesterol lowering effect of phytosterol nanoparticles.The results showed that phytosterol nanoparticles did not affect cell viability and had no cytotoxicity.They could effectively reduce the content of cholesterol in hypercholesterolemic cells and increase the activity of SOD and CAT,the antioxidant indexes in cells,and had a significant dose effect.But it had no significant effect on the bile acid content in the medium.These results indicated that phytosterol nanoparticles had good effects on lowering cholesterol and enhancing antioxidant activity of cells.
