摘要
目的探讨CD133与PANTR1在乳腺癌中的表达特征及其与细胞体外增殖和侵袭能力的关系。方法选取380例女性乳腺癌患者为研究对象,于术中取其癌组织(癌组织组)与癌旁正常组织(癌旁正常组织组),运用逆转录聚合酶链式反应(PT-PCR)法检测2组的CD133与PANTR1 mRNA相对量表达。同时取人乳腺癌(MCF-7)细胞进行培养,并划分为对照组、CD133过表达组、PANTR1过表达组、CD133抑制组以及PANTR1抑制组,探究各组细胞增殖与侵袭能力的差异。结果癌组织组的CD133与PANTR1 mRNA相对表达量[(0.023±0.002)、(0.018±0.002)]均高于癌旁正常组织组[(0.006±0.001)、(0.005±0.001)],有统计学差异(P<0.05)。CD133、PANTR1过表达组细胞增殖倍数[(10.54±1.45)、(10.26±1.13)]与侵袭细胞数[(43.88±3.69)个、(45.67±4.28)个]均高于对照组[(6.84±0.69)、(23.25±2.15)个],而CD133、PANTR1抑制组的细胞增殖倍数[(5.23±0.25)、(5.17±0.34)]与侵袭细胞数[(18.35±1.64)个、(18.13±1.22)个]均低于对照组[(6.84±0.69)、(23.25±2.15)个],有统计学差异(P<0.05)。结论CD133及PANTR1在乳腺癌组织内呈高表达,可增强乳腺癌细胞体外增殖与侵袭能力。
Objective To investigate the expression characteristics of CD133 and pantr1 in breast cancer and their relationship with cell proliferation and invasion in vitro.Methods 380 female patients with breast cancer who were diagnosed and treated were selected as the research objects.During the operation,the cancer tissues(cancer tissue group)and adjacent normal tissues(adjacent normal tissue group)were taken,and the relative expression of CD133 and pantr1 mRNA in the 2 groups was detected by reverse transcription polymerase chain reaction(PT-PCR);At the same time,human breast cancer(MCF-7)cells were taken and included in the control group,CD133 overexpression group,pantr1 overexpression group,CD133 inhibition group and pantr1 inhibition group respectively to explore the differences of cell proliferation and invasion in each group.Results The relative expressions of CD133 and pantr1 mRNA in the cancer group[(0.023±0.002),(0.018±0.002)]were significantly higher than those in the adjacent normal tissues[(0.006±0.001),(0.005±0.001)],with significant difference(P<0.05);The cell proliferation multiple[(10.54±1.45),(10.26±1.13)]and the number of invasive cells[(43.88±3.69),(45.67±4.28)]in the CD133 and pantr1 overexpression group were higher than those in the control group[(6.84±0.69),(23.25±2.15)]The cell proliferation multiple[(5.23±0.25),(5.17±0.34)]and the number of invasive cells[(18.35±1.64),(18.13±1.22)]in the pantr1 inhibition group were lower than those in the control group[(6.84±0.69),(23.25±2.15)],with significant difference(P<0.05).Conclusion CD133 and pantr1 are highly expressed in breast cancer,which can enhance the proliferation and invasion of breast cancer cells in vitro.
