摘要
依据酵母单细胞特性和细胞大小范围,采用改进的酵母菌分离方法对皇藏峪国家森林公园及其周边区域的酵母菌进行分离,基于26S rDNA D1/D2区序列对获得的酵母菌进行初步分类鉴定,调查可培养的酵母菌种资源.结果表明:增加滤膜过滤步骤能够减少丝状真菌和细菌的干扰,有利于酵母菌的分离;同一采样点不同寄生树种分离的酵母菌种类差异较大,菌种资源调查时要注意树种对调查结果的影响;鉴定的216株酵母菌株归属于37个属,68个种,其中子囊菌酵母25种,担子菌酵母43种;潜在新种11个,分属于子囊菌的Metschnikowia属和Wickerhamomyces属,担子菌的Cryptococcus属、Tremella属、Cystofilobasidium属、Rhodotorula属、Fibulobasidium属、Vishniacozyma属、Colacogloea属及Hamamotoa属.
According to the single cell characteristics and cell size range of yeast,using the improved method for yeasts isolation and culture,the yeasts in Huangcangyu National Forest Park and its surrounding areas were isolated.The obtained yeasts were preliminarily identified based on the gene sequence of 26S rDNA D1/D2 domain,and the culturable yeast strains were investigated.The results show that in this method the addition of membrane filtration step can reduce the interference of filamentous fungi and bacteria,and be beneficial to the separation of yeasts.The species of yeast isolated from different parasitic tree species at the same sampling point are quite different,so should pay attention to the influence of tree species on the investigation of yeasts.The isolated 216 yeast strains belong to 37 genera and 68 species,including 25 species of ascomycete yeast and 43 species of basidiomycete yeast.There are 11 potential new species,belonging to Metschnikowia and Wickerhamomyces of ascomycetes,Cryptococcus,Tremella,Cystofilobasidium,Rhodotorula,Fibulobasidium,Vishniacozyma,Colacogloea and Hamamotoa of basidiomycetes.
