芍仙萸颗粒对免疫性血小板减少症小鼠肝脾组织的影响

Effects of Shaoxianyu granules on liver and spleen in mice with immune thrombocytopenia

摘要 目的 探讨芍仙萸颗粒对免疫性血小板减少症(immune thrombocytopenia,ITP)小鼠肝脾组织及辅助性T细胞(helper T cell,Th cell)17/调节性T细胞(regulatory T cell,Treg cell)免疫平衡的影响。方法 将30只C57BL/6小鼠随机分为对照组、模型组、芍仙萸颗粒低剂量组(5.85 g/kg)、高剂量组(11.7 g/kg)和泼尼松组(5.4 mg/kg),每组6只。检测各组小鼠免疫相关指标、Th17和Treg细胞数,计算Th17/Treg比值,并观察小鼠肝脏、脾脏的形态变化。结果 与对照组相比,模型组小鼠血小板计数下降,血小板相关免疫球蛋白(platelet-associated immunoglobulin,PAIg)水平上升(P <0.05);芍仙萸颗粒低、高剂量组血小板计数上升,PAIg水平下降(P <0.05)。与对照组相比,模型组小鼠白介素(interleukin,IL)-17和转化生长因子(transforming growth factor,TGF)-β1水平下降,IL-10水平上升(P <0.05);芍仙萸颗粒低、高剂量组IL-17和TGF-β1水平上升,IL-10水平下降(P <0.05)。与对照组相比,模型组小鼠脾脏组织紊乱,白髓较大,红髓较小,髓索、髓窦内可见大量发育异常的巨核细胞,肝脏组织肿胀,肝小叶结构消失,汇管区及间质有大量炎症细胞浸润;芍仙萸颗粒低、高剂量组,肝脾组织病理改变较模型组显著改善,与泼尼松组疗效相当。与对照组相比,模型组小鼠Th17细胞数增加,而Treg细胞数减少,Th17/Treg比值上升(P <0.05);芍仙萸颗粒低、高剂量组,两者比值对应下降(P <0.05)。结论 芍仙萸颗粒可显著增加ITP小鼠血小板数量,有效控制出血,并通过调节Th17/Treg细胞免疫平衡对ITP小鼠的肝脾组织起到一定的保护作用。 Objective To investigate the effects of Shaoxianyu granules on liver and spleen tissue damage and helper T cell(Th)17/regulatory T cell(Treg) immune balance in mice with immune thrombocytopenia(ITP).Methods Thirty C57BL/6 mice were randomly divided into control group,model group,Shaoxianyu granules low-dose group(5.85 g/kg),high-dose group(11.7 g/kg) and prednisone group(5.4 mg/kg),6 mice in each group.The immune-related indicators,Th17 and Treg cells in each group were detected,the Th17/Treg ratio was calculated,and the morphological changes of the liver and spleen were observed.Results Compared with the control group,the platelet count decreased in the model group,and the concentration of platelet-related immunoglobulin(PAIg) increased(P<0.05).Compared with the control group,the platelet count increased and the concentration of PAIg decreased in Shaoxianyu granules low-dose and high-dose groups(P<0.05).Compared with the control group,the contents of interleukin(IL)-17 and transforming growth factor(TGF)-βdecreased in the model group,and the content of IL-10 increased(P<0.05),while the content of IL-17 and TGF-βincreased,and the content of IL-10 decreased in Shaoxianyu granules low-dose and high-dose groups(P<0.05).Compared with the control group,the spleen tissue of the mice in the model group was disordered,the white pulp was larger,the red pulp was smaller,a large number of abnormally developed megakaryocytes were seen in the medullary cord and sinus,the liver tissue was swollen,the hepatic lobule structure disappeared,and a large number of inflammatory cells infiltrated in the portal area and the interstitium.Compared with the model group,the pathological changes of the liver and spleen in Shaoxianyu granules low and high dose groups were significantly improved,and the curative effect was equivalent to that of the prednisone group.Compared with the control group,the number of Th17 cells in the model group increased,while the number of Treg cells decreased,and the ratio of Th17/Treg increased(P<0.05).Compared with the control group,the ratios of Th17/Treg in Shaoxianyu granules low and high-dose groups decreased correspondingly(P<0.05).Conclusion Shaoxianyu granules can significantly increase the number of platelets in ITP mice,effectively control bleeding,and protect the spleen and liver tissues of ITP mice by regulating the immune balance of Th17/Treg cells.
