吗啡通过miR-219-5p/NF-κB信号通路调控肺癌细胞增殖、迁移和侵袭

摘要 目的探讨吗啡对肺癌细胞增殖、迁移和侵袭的影响及其作用机制。方法不同浓度吗啡处理肺癌细胞A549,噻唑蓝(MTT)检测细胞增殖,Transwell小室法检测细胞迁移和侵袭,Western印迹检测细胞中基质金属蛋白酶(MMP)-2、MMP-9、细胞周期蛋白(Cyclin)D1、核因子(NF)-κB p65蛋白表达,实时荧光定量聚合酶链反应(qPCR)检测miR-219-5p表达。在A549细胞中转染miR-219-5p,或转染anti-miR-219-5p并使用25μmol/L吗啡处理,观察其对细胞增殖、迁移、侵袭及相关蛋白表达的影响。结果5、10、25μmol/L吗啡明显减少A549细胞存活率(P<0.05)。10、25μmol/L吗啡显著减少迁移细胞数、侵袭细胞数、MMP-2、MMP-9蛋白表达(P<0.05),并显著提高miR-219-5p表达(P<0.05)。高表达miR-219-5p明显减少A549细胞中CyclinD1、MMP-2、MMP-9表达量、细胞存活率、迁移细胞数和侵袭细胞数(P<0.05)。低表达miR-219-5p可以部分逆转吗啡对A549细胞增殖、迁移、侵袭、CyclinD1、MMP-2、-9、NF-κB p65蛋白表达的抑制作用。结论吗啡通过miR-219-5p/NF-κB信号通路抑制肺癌细胞增殖、迁移和侵袭。
