甘蔗居间分生组织腋芽的诱导研究

摘要 通过分析并解决甘蔗桂南亚08-186居间腋芽诱导的问题,为其他甘蔗品种提供理论基础;试验使用了MS基本培养基(蔗糖30 g/L+6-BA 0.1mg/L;pH 6.0)作为基础培养基添加不同浓度NAA(吲哚乙酸)和GA3(赤霉素),以找出最佳诱导组合;在培养至15天后,除了处理A1B1外其余均有腋芽冒出,2个处理A2B2、A2B3重复出芽瓶数分别为654和652;通过方差分析获MS+0.1mg/L NAA+0.05mg/L GA以及MS+0.1mg/L NAA+0.01mg/L GA较为理想的诱导桂南亚08-186居间分生组织腋芽培养基。本试验结果表明较低浓度的NAA配合较高浓度的GA3对桂南亚08-186居间分生组织具有较强的诱导分化作用。NAA可以打破分化居间分生组织的休眠,而GA3辅助促进了腋芽伸长和增高,二者相结合的方式可以打破休眠诱导分化居间分生组织长出腋芽。 The experiment used MS basic medium(sucrose 30 g/L+6-BA 0.1 mg/L;pH 6.0)as the base medium with different concentrations of NAA(indoleacetic acid)and GA3(gibberellin)to find out the best induction combination;After 15 days of incubation,all axillary buds emerged except for treatment A1B1,and the number of replicate buds in treatments A2B2 and A2B3 were 654 and 652,respectively;after comparing but contrasting the emergence,it was found that A2B3 had a neater emergence,therefore,A2B3(MS basic medium+0.1 mg/L NAA+0.05 mg/L GA3)was more ideal treatment for axillary bud induction.The results of this experiment showed that the lower concentration of NAA combined with higher concentration of GA3 had a strong effect on the induction of differentiation of the interstitial meristem of Guinanya08-186.NAA could break the dormancy of the differentiated interstitial meristem,while GA3 assisted in promoting the elongation and height of the axillary buds,and the combination of the two could break the dormancy and induce the axillary buds of the differentiated interstitial meristem.
