proBDNF与抑郁障碍认知功能损害的研究进展 被引量:2

摘要 为进一步了解抑郁障碍神经生物学机制并为未来的相关研究及诊疗提供新思路,对proBDNF与抑郁障碍认知功能损害的研究进行综述。抑郁障碍患者除了情绪障碍外还合并有认知功能损害,包括执行功能、记忆力、注意力和精神、运动技能等明显损害,抑郁障碍的发生不仅与神经递质、神经内分泌等功能障碍相关,并且一些细胞因子可以通过抑制BDNF诱导细胞凋亡而直接损伤神经细胞,相应脑区常出现BDNF表达降低以及功能下调,BDNF是抑郁障碍病理生理学中的关键因素,在BDNF中作为“惩罚信号”的proBDNF的积累可能促进抑郁障碍的发生,也会导致抑郁障碍认知功能损害。proBDNF抗体不仅可逆转抑郁样行为,还可改善认知功能,其有望成为一种可靠的生物学标记物进而提供一种更准确、快捷的诊断方式,基于此研发的新药有望改善双相情感障碍患者的预后,促进社会功能的恢复。 In order to further understand the neurobiological mechanism of depressive disorder and provide new ideas for relevant research and diagnosis and treatment in the future,this paper reviews the research on proBDNF and cognitive impairment of depressive disorder.In addition to emotional disorder,patients with depressive disorder are also complicated with cognitive impairment,including obvious impairment of executive function,memory,attention and psychomotor skills.The occurrence of depressive disorder is not only related to the dysfunction of neurotransmitters and neuroendocrine,but also some cytokines can directly damage nerve cells by inhibiting BDNF induced apoptosis.The expression and function of BDNF are often reduced in corresponding brain regions,BDNF is a key factor in the pathophysiology of depression.The accumulation of proBDNF as a"punishment signal"in BDNF may contribute to the occurrence of depression and the impairment of cognitive function of depression.ProBDNF antibody not only reversibly transformed depression like behavior,but also improved cognitive function.It is expected to become a reliable biomarker and provide a more accurate and fast diagnostic method.The new drugs developed based on this are expected to improve the prognosis of patients with bipolar disorder,promote the recovery of patients'social function and reduce the social burden.
