Lrrc15在斑马鱼胚胎血管发育中的作用

摘要 目的:探究Lrrc15基因在斑马鱼胚胎发育中的表达及其在血管发育中的作用。方法:采用全胚胎原位杂交技术检测Lrrc15基因在斑马鱼胚胎发育过程中的时空表达谱,显微注射吗啉修饰的反义寡核苷酸(morpholino oligos,MO)至内皮细胞被特异标记的转基因斑马鱼中下调Lrrc15的表达,利用共聚焦显微成像技术观察和分析胚胎血管发育的变化。结果:在受精后19、24、36和48 h均检测到Lrrc15基因在斑马鱼尾静脉丛的表达,受精后24 h表达最高。下调Lrrc15的表达导致斑马鱼尾静脉丛发育缺陷。结论:Lrrc15基因在斑马鱼胚胎发育中主要参与调控尾静脉丛形成,该基因的缺陷可导致尾静脉丛发育异常,提示Lrrc15基因在血管发育尤其是血管新生中的重要性,可为今后肿瘤治疗提供潜在的靶点。 Objective: To explore the expression and function of Lrrc15 during zebrafish vascular development.Methods: Firstly, the spatiotemporal expression of Lrrc15 during zebrafish embryo development through in situ hybridization(ISH) assay was examined. Then, Lrrc15 expression was made by injecting morpholino(MO) into the transgenic line Tg(fli1:EGFP) was knocked-down, in which the endothelial cells are labeled with the enhanced green fluorescent protein(EGFP). Finally, the confocal imaging technique was used to observe and analyze the dynamics of vascular development.Results: The expression of Lrrc15 was detected in the caudal vein plexus(CVP) region at 19, 24, 36 and 48 hours post fertilization(hpf), reaching the peak expression at 24 hpf. Confocal analysis revealed that downregulation of Lrrc15 resulted in development deficiency in CVP. Conclusions: Lrrc15 is required for vascular development in zebrafish, suggesting its role in tumorigenesis and progression. This discovery might offer potential target and strategy for cancer therapy.
