生血宝合剂对心衰大鼠心功能及MMP-2的影响

Effect of Shengxuebao Mixture on Cardiac Function and MMP-2 in Rats with Heart Failure

摘要 目的观察生血宝合剂对心衰大鼠心功能及基质金属蛋白酶2(MMP-2)的表达的影响。方法采用阿霉素腹腔注射法建立大鼠心衰模型,分为模型组、赖诺普利组、生血宝合剂组及空白对照组。给药6周后,行多普勒超声心动图检测各组大鼠左室舒张末内径(LVEDd)、左室收缩末期内径(LVEDs)、短轴缩短率(FS)、射血分数(EF);采用聚合酶链式反应(PCR)检测大鼠心肌组织中MMP-2的表达。结果与模型组比,生血宝合剂组LVEDd、LVEDs明显下降,FS、EF明显升高(P<0.05)。与模型组比,生血宝合剂组心衰大鼠心肌MMP-2的表达明显下降(P<0.05)。结论生血宝合剂可改善心衰大鼠心功能,抑制心肌组织中MMP-2的表达。 Objective To observe the effect of Shengxuebao mixture on heart function and expression of MMP-2 in rats with heart failure.Methods The heart failure model of rats was established by doxorubicin intraperitoneal injection and divided into model group,lisinopril group,Shengxuebao mixture group and blank control group.Six weeks after administration,left ventricular end diastolic diameter(LVEDd),left ventricular end systolic diameter(LVEDs),short axis shortening rate(FS)and ejection fraction(EF)were detected by Doppler echocardiography.PCR was used to detect the expression of MMP-2 in rat myocardium.Results Compared with model group,LVEDd and LVEDs in Shengxuebao mixture group were significantly decreased,FS and EF were significantly increased(P<0.05).Compared with model group,the expression of MMP-2 in heart failure rats in Shengxuebao mixture group was significantly decreased(P<0.05).Conciusion Shengxuebao mixture can improve the cardiac function of rats with heart failure and inhibit the expression of MMP-2 in myocardial tissue.
