摘要
为了解多酚氧化酶活性基因在黄淮麦区新育小麦品种中的分布,本研究利用其功能标记PPO18、F-8、PPO16和PPO_(2)9对96份供试材料的Ppo-A1、Ppo-B1和Ppo-D1位点的基因型进行分子检测,并探讨了这些基因型和基因型组合与多酚氧化酶活性值之间的关系。结果表明,在Ppo-A1位点上,基因型Ppo-A1a和Ppo-A1b在供试材料中的分布频率分别为66.7%和33.3%,以调控高多酚氧化酶活性的Ppo-A1a基因型分布频率较高;在Ppo-B1位点上,基因型Ppo-B1a和Ppo-B1b的分布频率分别为88.5%和11.5%,以Ppo-B1a基因型分布频率较高;在Ppo-D1位点上,基因型Ppo-D1a和Ppo-D1b的分布频率分别为68.8%和31.3%,以调控低多酚氧化酶活性的Ppo-D1a基因型分布频率较高。在Ppo-A1、Ppo-B1和Ppo-D1三个位点上,共检测到8个类型的基因型组合,其中调控低多酚氧化酶活性的Ppo-A1b/Ppo-B1a/Ppo-D1a基因型组合分布频率为17.7%,调控高多酚氧化酶活性的Ppo-A1a/Ppo-B1b/Ppo-D1b基因型组合分布频率为2.1%,其余6种调控中等多酚氧化酶活性的基因型组合分布频率合计为80.2%。本研究能够为小麦品质色泽改良提供一些信息。
In order to understand the distribution of the polyphenol oxidase genes in new bread wheat cultivars from Huanghuai wheat region,we applied the functional markers PPO18,F-8,PPO16 and PPO_(2)9 in 96 new bread wheat cultivars from Huanghuai wheat region to detect Ppo-A1,Ppo-B1 and Ppo-D1 sites allelic variation analysis,and discussesed the relationship between the variation and polyphenol oxidase activity values.In the Ppo-A1 locus,66.7%of the tested wheat varieties is Ppo-A1a and 33.3%of that is Ppo-A1b,and comparatively high polyphenol oxidase activity correlated with Ppo-A1 a genotype is higher;in the Ppo-B1 locus,88.5%of the tested wheat varieties is Ppo-B1a and 11.5%of that is Ppo-B1b,and the Ppo-A1a genotype is higher;In the Ppo-D1 locus,68.8%of the tested wheat varieties is Ppo-D1a and 31.3%of that is Ppo-D1b,and comparetively low polyphenol oxidase activity correlated with Ppo-D1a genotype is higher;there are eight genotype combinations in the Ppo-A1,Ppo-B1 and Ppo-D1 locus.The distribution frequency of the Ppo-A1b/Ppo-B1a/Ppo-D1a genotype combination regulating low polyphenol oxidase activity was 17.7%,the distribution frequency of the genotype combinations of Ppo-A1a/Ppo-B1b/Ppo-D1b that regulate the activity of high polyphenol oxidase was 2.1%,the total distribution frequency of the other six genotypes regulating the activity of polyphenol oxidase was 80.2%.This study provides important information for wheat flour color quality breeding.
