玉米叶夹角主效qLA7-1近等基因系构建 被引量:1

Construction of Maize Leaf Angle Major qLA7-1 Near-Isogenic Lines

摘要 叶夹角是植物重要的株型性状,近等基因系是研究其分子遗传机制的重要材料。本研究以紧凑型自交系‘廊黄’为供体亲本,平展型自交系‘TS141’为轮回亲本,构建了含有以‘TS141’为遗传背景的高代回交群体。通过对两个环境下两个亲本以及BC3F4进行表型鉴定及性状间相关分析发现,除总叶数外,BC3F4群体叶夹角与非轮回亲本‘廊黄’差异最小,叶夹角与叶向值均呈显著负相关(rE1=-0.549^(**),rE2=-0.337^(**))。利用该回交群体BC3F4,结合两对前景选择引物,得到含目标qLA7-1的11个单株,选取背景回复率大于80%的7个单株为叶夹角主效qLA7-1的目标单株,通过三个环境下表型鉴定,发现目标单株与非轮回亲本‘廊黄’的叶夹角差异不超过5%,目标单株的株高、穗位高、叶长、叶宽与轮回亲本‘TS141’相似度较高,7个目标单株可作为玉米叶夹角主效qLA7-1近等基因系。本研究为玉米叶夹角主效qLA7-1精细定位提供理论了基础。 Leaf angle is an important plant morphology of plants,and near-isogenic lines are important materials for studying their molecular genetic mechanisms.In this study,the compact inbred line’Langhuang’was used as the donor parent,and the flat inbred line’TS141’was used as the recurrent parent.A high-generation backcross population with the genetic background of’Langhuang’was constructed.Environmental phenotyping and correlation analysis between traits showed that,except for the total number of leaves,the difference between the leaf angle of the BC3F4 population and the non-recurrent parent’Langhuang’was the smallest,and the leaf angle and leaf orientation values were both significantly negatively correlated(rE1=-0.549^(**),rE2=-0.337^(**)).Using this backcross population,combined with two pairs of foreground selection primers,11 individual plants containing the target qLA7-1 were obtained,and 7 individual plants with a background recovery rate greater than 80%were selected as the target individual plants with leaf angle main effect qLA7-1.Through three environmental phenotypic identification,the difference in leaf angle between the target individual plant and the non-recurring parent’Langhuang’does not exceed 5%,and the target individual plant differs in plant height,ear height,leaf length,and leaf width.The parental’TS141’has a high similarity,and 7 target individual plants can be used as maize leaf angle major effect qLA7-1 near-isogenic lines.This study provided a theoretical basis for the fine positioning of main effect qLA7-1 of maize leaf angle.
