摘要
目的:探讨银屑病患者外周血微小RNA(miR)-146a、皮损部位黑素瘤分化相关基因(MDA)5、白细胞介素(IL)-6、Ⅰ型干扰素(IFN)-Ⅰ水平与银屑病皮损面积和严重程度指数(PASI)评分的相关性。方法:选择该院2019年8月—2021年5月就诊的30例银屑病患者为银屑病组,将30例整形科诊治的正常人为对照组。采用聚合酶链式反应(PCR)检测2组miR-146a表达。采用实时荧光定量PCR(RT-qPCR)和免疫印迹法(western blot)检测2组MDA5、IL-6及IFN-Ⅰ的表达。银屑病组患者皮损MDA5、miR-146a、IL-6及IFN-Ⅰ表达与PASI评分的相关性采用Spearman相关性分析。应用受试者工作特征曲线(ROC)检验MDA5、IL-6及IFN-Ⅰ对银屑病患者皮损的评估价值。结果:正常对照组MDA5、miR-146a、IL-6及IFN-ⅠmRNA表达水平均低于银屑病组,差异有统计学意义(P<0.05)。正常对照组MDA5、IL-6、IFN-Ⅰ蛋白表达均低于银屑病组,差异均有统计学意义(P<0.05)。Spearman相关性分析结果示MDA5、miR-146a、IL-6及IFN-Ⅰ水平与银屑病患者PASI评分均呈正相关(P<0.05)。ROC曲线显示MDA5、IL-6及IFN-Ⅰ的曲线下面积分别为0.901、0.952、0.962,结果均有统计学意义(P<0.05)。结论:银屑病患者外周血miR-146a、MDA5及其下游IL-6及IFN-Ⅰ表达水平与PASI评分均有相关性。
Objective:To investigate the correlation of peripheral blood microRNA-146a(miR-146a),melanoma differentiationassociated gene 5(MDA5),interleukin-6(IL-6)and interferon typeⅠ(IFN-Ⅰ)levels with psoriasis lesion area and severity index(PASI)scores in patients with psoriasis.Methods:Thirty patients with psoriasis treated in our hospital from August 2019to May 2021 were selected as the psoriasis group,and 30 healthy people from the plastic department were included in the normal control group.Polymerase chain reaction(PCR)was used to detect the expression of miR-146a in the two groups.Realtime fluorescence quantitative PCR(RT-qPCR)and immunoblotting(western blot)were used to detect the expression of MDA5,IL-6 and IFN-Ⅰin the two groups.The correlation between the expressions of MDA5,miR-146a,IL-6 and IFN-Ⅰin skin lesions and PASI scores of the psoriasis group were analyzed by Spearman correlation analysis.The subject operating characteristic curve(ROC)was applied to analyze the value of MDA5,IL-6,and IFN-Ⅰfor the assessment of skin lesions in patients with psoriasis.Results:The mRNA levels of MDA5,miR-146a,IL-6,IFN-Ⅰin normal control group were lower than those in the psoriasis group(P<0.05)The protein expressions of MDA5,IL-6 and IFN-Ⅰin the normal control group were lower than those in the psoriasis group,with statistically significant difference(P<0.05).Spearman correlation analysis showed that MDA5,miR-146a,IL-6,IFN-Ⅰlevels were positively correlated with the PASI score in patients with psoriasis(P<0.05).ROC curve showed that the area below the line of MDA5,IL-6 and IFN-Ⅰwas 0.901,0.952 and 0.962,respectively,and the results were all statistically significant(P<0.05).Conclusion:The expression levels of miR-146a,MDA5 and its downstream IL-6 and IFN-Ⅰin peripheral blood of patients with psoriasis were correlated with PASI scores.
