摘要
“卢桉1#”具有速生丰产、抗寒抗病等特点,为推广该无性系在造林中的应用,文章对其无性系组培快繁技术进行了研究。结果表明:较适宜的茎段外植体诱导培养基为MS+6BA1.0 mg·L^(-1)+NAA0.2 mg·L^(-1)+糖30 g·L^(-1)+卡拉胶5.6 g·L^(-1);较适宜增殖继代培养基为改良MS+6BA0.5~0.6 mg·L^(-1)+NAA0.1 mg·L^(-1)+糖30 g·L^(-1)+卡拉胶5.5 g·L^(-1);较适宜的不定根诱导培养基为0.5MS+ABT1#2.0 mg·L^(-1)+IBA0.5 mg·L^(-1)+糖15~20 g·L^(-1)+卡拉胶6.5 g·L^(-1)。
Eucalyptus clone of Lu’An 1#has the characteristics of rapid growth,high yield,cold resistance and disease resistance.In order to promote its application in afforestation,tissue culture and rapid propagation of this clone was examined in this study.The results showed that the optimal medium for induction of stem explants was MS+6BA1.0 mg·L^(-1)+NAA0.2 mg·L^(-1)+sugar 30 g·L^(-1)+carrageenan 5.6 g·L^(-1),while the optimal medium for proliferation and subculture was MS+6BA0.5~0.6 mg·L^(-1)+NAA0.1 mg·L^(-1)+sugar30 g·L^(-1)+carrageenan 5.5 g·L^(-1).Meanwhile,the optimal medium for adventitious root induction was 0.5MS+ABT1#2.0 mg·L^(-1)+IBA0.5 mg·L^(-1)+sugar15~20 g·L^(-1)+carrageenan 6.5 g·L^(-1).
