产褐藻胶裂解酶弧菌HB161653的产酶条件优化 被引量:1

摘要 为提高弧菌HB161653产褐藻胶裂解酶活性,该研究通过单因素和正交试验对弧菌HB161653的产酶条件进行了优化。结果表明,最优的发酵培养基组成为海藻酸钠5 g/L,蛋白胨7.5 g/L,NaCl 25 g/L,KHPO0.2 g/L,Mg SO0.1 g/L;最优发酵条件为培养基初始pH 7.0,接种量1.5%,发酵温度28℃,发酵周期12 h。在此优化产酶条件下,褐藻胶裂解酶活力可达42.1 U/mL,较优化前(26.0 U/mL)提高了62%。该研究可为弧菌HB161653产褐藻胶裂解酶的规模化生产和褐藻寡糖的制备提供理论依据。 In order to improve the activity of alginate lyase produced by Vibrio sp.HB161653,the enzyme production conditions were optimized by single-factor and orthogonal tests.The results showed that the optimal fermentation medium was as follow:sodium alginate 5 g/L,peptone 7.5 g/L,Na Cl 25 g/L,K2HPO40.2 g/L,Mg SO40.1 g/L,and the optimal fermentation conditions were initial pH of the medium 7.0,inoculum 1.5%,temperature 28℃,and time 12 h.Under these optimized enzyme production conditions,the activity of alginate lyase reached 42.1 U/ml,which was 62%higher than that before optimization(26.0 U/ml).This study provided a theoretical basis for the large-scale production of alginate lyase produced by Vibrio sp.HB161653 and the preparation of alginate oligosaccharides.
