快速测定唐古特白刺种子生活力的研究

摘要 【目的】快速评价唐古特白刺种子的发芽潜力。【方法】对浸种温度、浸种时间、TTC溶液浓度、染色温度和染色时间进行5因素4水平正交试验以测定唐古特白刺种子的生活力,并进行回归分析。【结果】浸种温度、浸种时间、TTC溶液浓度、染色温度和染色时间5个因素都是影响唐古特白刺种子生活力测定的主要因素,其中浸种时间影响最大,浸种温度与TTC浓度次之;试验测得种子生活力的最佳检测条件是35℃下浸种20 h、0.3%TTC染色液、25℃恒温、黑暗条件下染色15 h,84%的种子表现出强生活力;拟合方程可以预测试验过程中76.8%的情况。【结论】研究筛选出的最优组合可以快速测定唐古特白刺种子的生活力,并且回归方程可以检测试验中出现的异常值,可为后续育苗造林提供参考。 【Objective】The studt aimed to evaluate quickly the germination potential of Nitraria tang⁃utorum seeds.【Method】Using N.tangutorum seeds as material,a 5-factor 4-level orthogonal test was performed on the seed soaking temperature,seed soaking time,TTC solution concentration,dyeing temperature and dyeing time to determine the seed viability,and regression analysis was performed.【Result】A total of 5 factors are the main factors that affect the determination of seed viability,among which the time of soaking seeds has the greatest influence,followed by soaking temperature and TTC concentration.The best detection conditions are soaking seeds at 35℃for 20 hours,0.3%TTC staining solution,25℃constant temperature,and staining for 15 hours under dark conditions.The seeds about 84%showed strong viability,the fitting equation can predict 76.8%of the conditions during the experiment.【Conclusion】The optimal combination screened out in this study can quickly measure the viability of a batch of N.tangutorum,and the regression equation can detect abnormal values in the experiment,providing a reference for subsequent seedling cultivation and afforestation.
